姜黄素联合多烯紫杉醇诱导PC-3细胞凋亡的实验研究

摘    要:目的 探讨姜黄素联合多烯紫杉醇对人前列腺癌PC-3细胞凋亡的影响及其作用机制.方法 将体外培养的PC-3细胞分为对照组(仅加培养基)、姜黄素组(5.0 μmol/L姜黄素处理)、多烯紫杉醇组(2.5 nmol/L多烯紫杉醇处理)及联合组(5.0 μmol/L姜黄素与2.5 nmol/L多烯紫杉醇联合处理),作用24 h.用流式细胞仪分析PC-3细胞凋亡率,逆转录聚合酶链反应(RT-PCR)和Western blot法分别检测Bcl-2及Bax mRNA和蛋白的表达情况.结果 姜黄素和多烯紫杉醇均可诱导PC-3细胞凋亡,二者联合用药组的细胞凋亡率明显高于单药组(P<0.05);RT-PCR及We

Abstract:Objective To investigate the effects of curcumin combined with docetaxel on apoptosis of human prostate cancer PC-3 cells and its mechanism.Methods Cultured PC-3 cells were divided into control group(only medium added),curcumin group(treatment with 5.0 μmol/L curcumin),docetaxel group(treatment with 2.5 nmol/L docetaxel) and combination group(combination treatment of 5.0 μmol/L curcumin and 2.5 nmol/L docetaxel),and were treated for 24 hours.Flow cytometry was employed to analyze the apoptotic rate of PC-3 cells,reverse transcriptase-polymerase chain reaction(RT-PCR) and Western blot were used to detect the mRNA and protein expression of Bcl-2 and Bax,respectively.Results Both curcumin and docetaxel could induce apoptosis in PC-3 cells,the apoptotic rate of cells in combination group was higher than that in monotreatment groups(P<0.05).RT-PCR and Western blot demonstrated that both curcumin and docetaxel down-regulated mRNA and protein expression of Bcl-2 and up-regulated mRNA and protein expression of Bax,and changes of Bax and Bcl-2 expression in combination group were more significant.Conclusion Curcumin and docetaxel can synergistically induce apoptosis in PC-3 cells via down-regulating Bcl-2 and up-regulating Bax.
