宫颈癌组织的MNX1-AS1水平及临床意义

摘    要:目的探讨宫颈癌组织的MNX1反义RNA 1(MNX1-AS1)水平及临床意义。方法采用实时定量PCR检测本院2017年9月至2019年6月手术切除的107例宫颈癌组织和98例正常宫颈组织的MNX1-AS1水平,采用受试者工作特征(ROC)曲线评估组织MNX1-AS1水平诊断宫颈癌的效能,进一步分析MNX1-AS1水平与宫颈癌临床病理特征的关系。GEPIA分析癌症基因组图谱(TCGA)数据库中宫颈癌组织MNX1-AS1水平与预后的关系。结果宫颈癌组织的MNX1-AS1水平为2.847±1.076,高于正常宫颈组织的1.182±0.712(P<0.05);ROC曲线结果显示组织MNX1-AS1水平诊断宫颈癌的曲线下面积为0.906(95%CI:0.867~0.945),当以1.634为截断点时对应的约登指数最大,灵敏度和特异度分别为85.98%和79.59%。MNX1-AS1水平与宫颈癌患者的年龄、月经状态、肿瘤直径、病理类型和分化程度无关(P>0.05),而与FIGO分期、浸润深度和淋巴结转移有关(P<0.05),其中FIGO分期Ⅲ~Ⅳ期、T3~T4期及淋巴结转移的MNX1-AS1水平分别为3.188±0.907、3.427±0.817和3.219±0.902,均高于Ⅰ~Ⅱ期、浸润深度T1~T2期及淋巴结未转移的2.590±1.127、2.426±1.050和2.171±1.046(P<0.05)。GEPIA在线分析显示MNX1-AS1水平与宫颈癌的总生存期和无病生存期均有关,其中MNX1-AS1高水平的预后较差风险分别是低水平的2.2和1.9倍(P<0.05)。结论宫颈癌组织的MNX1-AS1呈高表达,与宫颈癌的病情进展和预后有关,可能促进了宫颈癌病情进展,有作为宫颈癌诊治的潜能。

Abstract:Objective To explore the level and clinical significance of MNX1 antisense RNA 1(MNX1-AS1)in cervical cancer tissues.Methods Real-time quantitative PCR was used to detect the MNX1-AS1 levels in 107 cervical cancer tissues and 98 normal cervical tissues from September 2017 to June 2019.Receiver operating characteristic(ROC)curve was used to evaluate the efficacy of MNX1-AS1 in the diagnosis of cervical cancer,and relationship between MNX1-AS1 level and clinicopathological characteristics of cervical cancer was analyzed.GEPIA was used to analyze the relationship between MNX1-AS1 level and prognosis of cervical cancer in The Cancer Genome Atlas(TCGA)database.Results The MNX1-AS1 level in cervical cancer tissues was 2.847±1.076,higher than 1.182±0.712 of normal cervical tissues(P<0.05).The ROC results showed that the area under the curve of MNX1-AS1 was 0.906(95%CI:0.867-0.945).When 1.634 was chosen as the truncation point,the corresponding Jordan index was the largest with the sensitivity of 85.98%and specificity of 79.59%.The MNX1-AS1 level was not related to age,menstrual state,tumor diameter,pathological type and differentiation degree of cervical cancer patients(P>0.05),but related to FIGO stage,invasion depth and lymph node metastasis(P<0.05).MNX1-AS1 levels in tissues of FIGO stageⅢ-Ⅳ,invasion depth T3-T4 and lymph node metastasis were 3.188±0.907,3.427±0.817 and 3.219±0.902,higher than 2.590±1.127,2.426±1.050 and 2.171±1.046 in tissues of FIGO stageⅠ-Ⅱ,invasion depth T1-T2 and without lymph node metastasis(P<0.05).GEPIA online analysis showed that the MNX1-AS1 level was related to disease-free survival and overall survival of cervical cancer,and the risk ratios of poor prognosis of high MNX1-AS1 level were 2.2 and 1.9 folds of low level(P<0.05).Conclusion The high expression of MNX1-AS1 in cervical cancer is related to the progression and prognosis of cervical cancer,which may promote the progression of cervical cancer,and has the potential of diagnosis and treatment of cervical cancer.
