脑络欣通对脑缺血再灌注损伤模型鼠神经细胞凋亡及相关基因的影响

摘    要:目的:研究脑络欣通对脑缺血再灌注半暗带神经细胞凋亡及相关基因表达的影响。方法:采有气虚血瘀局灶性脑缺血再灌注模型鼠,大脑中动脉阻断2h,再灌注ld、3d,用TUNEL法和免疫组化染色法分别检测缺血半暗带凋亡细胞和凋亡相关基因bcl-2、bax蛋白表达。结果:与模型组比较,脑络欣通组缺血半暗带的凋亡细胞显著减少,bcl-2蛋白表达显著增加,bax蛋白表达显著减少。结论:脑络欣通可能通过促进bcl-2基因的表达,抑制bax基因的表达对半暗带的细胞凋亡产生抑制作用。

Abstract:Objective:To investigate the effects of Naoluoton g on apoptosis and relating gene in local cerebral ischemic reperfusive model ra ts of Qi deficiency and Blood stasis.Methods:The middle cerebral arteries (MCA) of SD rats were occluded for 2 hours and reperfused for 1-3 days.Apoptosis was d etected by teminal deoxynucleotidyl transferase\|mediated dUTP nick end\|labelin g(TUNEL).Bcl-2 and bax expression was detected by immunohistochemistry method.R esults:In the Naoluotong groups,the apoptotic cells were significantly decreased ( P <0.01),bcl-2 protein expression was significantly increased( P <0.01) .Bax pro tein expression was significantly decreased.Conclusion:Naoluotong can prevent ap optosis in local cerebral ischemic reperfusive model rats due to the emhanced ex pression of bcl-2 and decreased expression of bax.
