摘    要:目的 研究心肌缺血预适应时一氧化氮-环磷酸鸟苷(NO-cGMP)通路的变化,并探讨引起变化的原因及意义.方法 选择SD大鼠,随机分成假手术组(CON组,n=6),预适应组(IP组,n=12),缺血再灌注组(I/R组,n=12)和预适应程序组(PC组,n=36).以手术套管法造成左冠状动脉主干缺血再灌注,损伤后取心脏组织检测NO、一氧化氮合成酶(NOS)和cGMP水平.结果 IP组NO、NOS和cGMP较I/R组明显增高(P<0.05);PC组中,NO及cGMP含量随缺血及再灌注呈周期性波动.结论 NO-cGMP通路参与预适应的保护作用,并可能是预适应作用的激发者和中介者.

Abstract:Objective To study the changes of nitric oxidecGMP pathway during ischemic preconditioning(IP) and discuss the reasons and implication.Methods SD rats were randomly divided into 4 groups: sham-operation group(n=6),IP group(n=12),ischemia/reperfusion(I/R) group(n=12) and PC group(n=36).Surgical procedures included intermittent left coronary artery occlusion and reperfusion.After the procedure,the hearts were extracted.The NO content and NOS activity were analyzed by the method of biochemistry,and the content of cGMP was determined by radioimmunoassay(RIA).Results The content of NO and cGMP,and the activity of NOS were much higher in IP group than those in I/R group(P<0.05).In PC group,the contents of NO and cGMP periodically varied with ischemia and reperfusion ischemia.Conclusion NO-cGMP pathway takes part in the protective effect of ischemic preconditioning.The increased contents of NO and cGMP,and the fluctuation of them during the preconditioning protocols show that NO and cGMP are probably the mediators and triggers of ischemic preconditioning.
