大鼠肝微粒体细胞色素b5对细胞色素P450在体温调节中作用影响的探讨

摘    要:目的 :通过观察发热、降温和解热时大鼠肝微粒体细胞色素b5(CYb5)含量的变化 ,探讨CYb5对细胞色素P 4 5 0 (CYP 4 5 0 )在体温调节中的作用。方法 :连续监测大鼠体温 ,并用改进的木村和佐藤方法测量第 1次注射不同制剂后 2 4h肝微粒体CYb5含量。结果 :腹腔注射脂多糖后大鼠体温升高 (P <0 .0 5 ) ,注射后 2 4hCYb5含量下降 (P <0 .0 5 ) ;注射丁香酚后大鼠体温下降 (P <0 .0 5 ) ,注射后 2 4hCYb5含量增加 (P <0 .0 5 ) ;腹腔注射脂多糖后 3h再腹腔注射丁香酚大鼠体温先升高然后恢复 ,注射后 2 4hCYb5含量与对照组相比无统计学意义。结论 :随着注射不同试剂大鼠体温变化的同时 ,CYb5含量变化方向与CYP 4 5 0含量方向相同 ,表明CYb5可能影响CYP 4 5 0在体温调节中的作用

Abstract:Objective: Our aims were to investigate change of content of cytochrome b 5(CYb 5) in the hepatocellular microsome in rats when febrile,hypothermal,and antipyretic, and to discuss the effects of CYb 5 on the role of cytochrome P450(CYP450) in regulation of body temperature.Methods:We measured the rats' temperature with digital thermometer and the content of CYb 5 in the hepatocellular microsome with modified Omura and Sato method.Results:The rats' temperature increased(P<0.05) after injection with lipopolysaccharide(LPS) and the content of CYb 5 decreased(P<0.05). The rats' temperature decreased(P<0.05) after injection with eugenol and the content of CYb 5 increased(P<0.05). In the LPS/eugenol group, the rats' temperature increased at first(P<0.05) and then restored. Change of the content of CYb 5 24 hours after the first injection was not statistical different from the control group.Conclusion:When the rats' temperature changed after different injections the content of CYb 5 changed in the same way with the change of the content of CYP450,wiich means that CYb 5 may affect the role of CYP450 in regulation of body temperature.
