淫羊藿苷对人牙周膜细胞增殖及骨向分化的影响

摘    要:目的:探讨淫羊藿苷对人牙周膜细胞增殖及骨向分化的影响,为淫羊藿苷在治疗牙周病方面的应用提供一定依据。方法:体外培养人牙周膜细胞,矿化诱导条件下将第3代细胞与10~0.001mg/L,6个浓度的淫羊藿苷作用,通过噻唑蓝(MTT)比色法、ALP活性测定及BSP表达水平,反应其对人牙周膜细胞增殖及分化的影响。结果:作用24h,各药物浓度均能促进人牙周膜细胞增殖(P〈0.05)。作用48h,1~0.001mg/L组可促进人牙周膜细胞增殖(P〈0.05)。作用72h,0.1~0.001mg/L组可促进人牙周膜细胞增殖(P〈0.05),10mg/L组抑制人牙周膜增殖(P〈0.05);作用72h,1~0.001mg/L组可促进人牙周膜碱性磷酸酶(ALP)活性(P〈0.05);作用72h,0.1~0.001mg/L组的BSP表达水平较对照组显著增加(P〈0.05)。结论:淫羊藿苷在一定浓度范围内可促进人牙周膜细胞增殖及骨向分化。

Effects of Icariin on the Proliferation and Osteogenic Differentiation of Human Periodontal Ligament Cells

Abstract:Objective: To investigate the effects of icariin on the proliferation and osteogenic differentiation of human periodontal ligament cells in order to provide a basis for the application of icariin in periodontal disease treatment.Methods: Human periodontal ligament cells were cultured in vitro.Under the conditions of mineralization induce,the third passage of human periodontal ligament cells were treated with 6 different concentrations(10-0.001μg/ml) of icariin.MTT colorimetric assay,ALP and BSP were performed to assess the effects of icariin on the proliferation and osteogenic differentiation.Results: All concentrations of icariin promoted proliferation of human periodontal ligament cells for 24 hours(P0.05).1-0.001μg/ml icariin promoted proliferation of human periodontal ligament cells for 48 hours(P0.05).0.1-0.001μg/ml icariin promoted proliferation of human periodontal ligament cells for 72 hours(P0.05),while 10μg/ml icariin inhibited proliferation of human periodontal ligament cells(P0.05).1-0.001μg/ml icariin improved ALP activity expression for 72 hours(P0.05).Compared with the control group,BSP expression of 0.1-0.001μg/ml cariin were increased significantly for 72 hours(P0.05).Conclusion: Using appropriate icariin could promote proliferation and osteogenic differentiation of human periodontal ligament cells.
