猪下腔静脉滤器置入术后MMP-2、MMP-9表达变化及意义

目的观察猪下腔静脉滤器置入术后内膜和中膜的变化以及下腔静脉管壁中MMP-2、MMP-9的表达分布及变化,并探讨其意义。方法将12头广西巴马小型猪随机分为对照组和3组实验组,每组3头,实验组分别在置入下腔静脉滤器后7天、28天和56天取出置入下腔静脉滤器段的下腔静脉,采用HE染色方法观察内膜及中膜的变化,免疫组织化学染色和Western blot观察分析MMP-2,MMP-9的表达分布及变化。结果下腔静脉中膜在滤器置入后7天时较对照组明显增厚,28天及56天逐渐萎缩。内膜在7天时较对照组无明显增生,28天及56天开始围绕滤器明显增生,且56天内膜增生程度明显比28天严重。下腔静脉壁中MMP-2、MMP-9在滤器置入后7天时表达水平达到峰值,28天及56天表达水平逐渐下降(P<0.05)。MMP-2与MMP-9的表达变化与中膜变化呈正相关。结论MMP-2、MMP-9表达升高可能与下腔静脉滤器置入后中膜早期增殖的细胞向内膜迁移形成新生内膜有关。

Expression and Significance of MMP-2 and MMP-9 After Inferior Vena Cava Filter Implantation in Pigs

Objective To observe the changes of intima and media after implantation of pig inferior vena cava filter and the expression and distribution of MMP-2 and MMP-9 in the wall of inferior vena cava tube,and explore its significance.Methods Twelve Guangxi Bama mini-pigs were randomly divided into a control group and three experimental groups,with 3 animals in each group.The experimental group had inferior vena cava removed at 7 days,28 days,and 56 days after placement of the inferior vena cava filter,respectively.In the inferior vena cava of the filter segment,HE staining was used to observe the changes of intima and media.Immunohistochemical staining and Western blot analysis were used to analyze the expression distribution and changes of MMP-2 and MMP-9.Results The inferior vena cava media was significantly thicker than the control group at 7 days after the filter was placed,and gradually atrophied at 28 and 56 days.The intimal membrane had no obvious hyperplasia at 7 days compared with the control group.At 28 and 56 days,there was obvious hyperplasia around the filter and the degree of hyperplasia of the membrane was significantly more severe than that at 28 days.The expression levels of MMP-2 and MMP-9 in the inferior vena cava wall reached a peak at 7 days after the filter was placed,and the expression levels gradually decreased at 28 and 56 days(P<0.05).The change of MMP-2 and MMP-9 expression was positively correlated with the change of media.Conclusion The increased expression of MMP-2 and MMP-9 may be related to the migration of cells proliferating into the intima and forming a new intima after inferior vena cava filter insertion.
