芹菜素诱导人肝癌Hep G2细胞分化

摘    要:目的:研究芹菜素诱导人肝癌Hep G2细胞分化作用。方法:体外培养人肝癌Hep G2细胞,瑞氏-姬姆萨和考马斯亮蓝染色观察Hep G2细胞形态和微管蛋白排列的变化。放射免疫法检测细胞液甲胎蛋白(Alpha-fetoprotein,AFP);酶促反应试剂盒检测细胞碱性磷酸酶(alkaline phosphatase,ALP)和γ-谷胺酰转肽酶(γ-glutamyltranspeptidase,γ-GT)的活性;Diamondstone分光光度法测定细胞酪氨酸α-酮戊二酸转氨酶(tyrosine-α-Ketoglutaric acid transaminase,TAT)的含量。结果:10μmol/L芹菜素和全反式维甲酸处理人肝癌Hep G2细胞48小时后,细胞的形态和微管微丝由肿瘤细胞向成熟细胞分化;细胞AFP的分泌和γ-GT的合成显著降低(P<0.01);ALP和TAT的合成则显著升高(P<0.01)。结论:全芹菜素具有诱导人肝癌Hep G2细胞分化作用。

Abstract:Objective To investigate the effect of apigenin on the induction of differentiation of human primary hepatocarcinoma cell strain Hep G2.Methods The Hep G2 cells were cultured in vitro,Gimsa staining and Coomassie brilliant blue staining were used to observe the alteration of cellular morphology and tubulin arrangement of Hep G2 cells.The Alpha-fetoprotein(AFP)secretary amount of the cells were assayed by radioimmunoassay(RIA).The activity of alkaline phosphatase(ALP)and γ-glutamyltranspeptidase(γ-GT)were tested by enzymatic reaction kit.Diamondstone spectrophotometry was employed to determine the synthese of tyrosine-α-Ketoglutaric acid transaminase(TAT).Results After treatment with apigenin(Api)and all-trans retinotic Acid(RA)at 10 μmol/L for 48h,Hep G2 cells were differentiated from tumour cells to mature cells,based on the observation of alteration of cellular morphology and tubulin arrangement of Hep G2 cells.The synthesis of TAT and ALP in Hep G2 cells were increased significantly(P<0.01),nevertheless the secretory amount of AFP and the specific activities of γ-GT were decreased distinctly(P<0.01).Conclution Apigenin can induce the differentiation of human primary hepatocacinoma Hep G2 cells.
