CD40分子在胃癌组织中的表达及其与细胞凋亡的相关性

目的研究CD40分子表达与胃癌浸润转移等临床病理的关系,并探讨其与胃癌细胞凋亡之间的相关性。方法应用免疫组化技术检测56例胃癌组织和32例癌旁正常组织中CD40分子表达,采用TUNEL法检测肿瘤细胞的凋亡情况。结果CD40分子在胃癌组织中的阳性表达率为33.9％,明显高于癌旁正常组织(P<0.01);CD40阳性表达与淋巴结转移及远处转移有关(P<0.01),与肿瘤分化程度、浸润深度和瘤块大小均无关。CD40阳性组凋亡指数(0.74±0.23)与阴性组(1.48±0.52)相比有显著性差异(P<0.01)。结论CD40表达与胃癌转移、凋亡生物学行为密切相关,有可能作为胃癌诊断、评价转移及预后的客观指标之一,并有望成为胃癌生物治疗的靶分子之一。

Expression of CD40 and its relationship with apoptosis in gastric cancer

Objective To investigate the relationship between CD40 expression and clinical pathologic factors of gastric cancer, and to explore the correlation of CD40 with apoptosis in gastric cancer. Methods The expression of CD40 was examined by immunohistochemistry in human gastric cancer tissues(56 cases) and adjacent nomal gastric tissues(32 cases). Apoptosis of the gastric cancer cell was detected by TUNEL. Results The immunoreactivity of CD40 increased in 33. 9% (19/56) gastric cancer tissues, which was significantly higher than that in tumor-free tissues (6. 3%) (P< 0. 01). A statistically significant correlation was found between the expression of CD40 and the meta-static spread of the tumor(P<0. 01). CD40 expression showed no significant relationship with tumor size and histological types. The expression of CD40 was inversely correlated with apoptosis index(AI) (P<0. 01). Conclusion The results suggest that CD40 signaling may play a role in apoptosis and me-tastatic spread of gastric cancer. Measurement of CD40 expression in gastric cancer may be useful in diagnosis, evaluation of lymph node metastasis and prognosis.
