摘    要:目的 建立一种稳定、快速的柱前衍生-反相高效液相色谱测定血清和脑脊液中多种氨基酸的方法。方法 样品用ACCQ Fluor试剂进行衍生化后,在C18色谱柱上采用三梯度淋洗分离,荧光检测器检测。结果 本方法回收率在101.3%-97.9%之间,10次测定的RSD均小于5%。结论 1个样品在36min内可完成17种氨基酸的分离测定,各种氨基酸之间分离良好,方法快速、稳定、准确,可用于临床及科研样品的检测。

Abstract:Objective To establish a method to determine amino-acid in CSF and serum by HPLC after pre-column derivatization.Methods The serum and CSF sample were derived by ACCQ.Fluor reagent were separated by three-gradient elution and detected by Fluorscence detector.Results The 17 amino-acid were separated completely.The method recovery were between 97.9% and 101.3% with the RSD were less than 5%.Conclusion The method are used to determine 35 CSF and 20 serum from patient,which showed that the method can be applied in clinical detection and science research.
