人参皂甙Rg1与坐骨神经损伤后修复相关性研究

摘    要:目的:探讨中药人参皂苷Rg1(GsRg1)对大鼠坐骨神经损伤后恢复作用以及是否有局部释放神经生长因子的参与。方法于2013年5月购买健康雄性Wistar大鼠,2月龄,共270只,随机均分为6组,每组45只。离断大鼠右侧坐骨神经后,立即在显微镜下缝合,左侧坐骨神经为空白对照,手术后,大鼠随机分为生理盐水组、GsRg12 mg 、4 mg、8 mg、12 mg组和甲钴胺组,分别腹腔内注射,手术2、4、8周后,通过称量大鼠双侧小腿三头肌计算其湿重比率,据此评价坐骨神经再生和功能恢复情况;采用免疫组织化学和Western blot技术检测大鼠坐骨神经NGF表达的变化。结果①小腿三头肌湿重比,各GsRg1剂量组及甲钴胺组也明显高于生理盐水组, GsRg1组中4 mg组、8 mg组高于GsRg1其它2组及甲钴胺组,GsRg12 mg组、12 mg组与甲钴胺组小腿三头肌湿重比恢复相似。②NGF免疫组化及Western blot结果显示,GsRg1各组及甲钴胺组NGF表达明显高于生理盐水组。GsRg14 mg和GsRg18 mg组NGF表达多于甲钴胺组、GsRg12 mg组、12 mg组。甲钴胺组与GsRg12 mg组、12 mg组NGF表达相似。结论①GsRg1剂量为4 mg、8 mg的促神经再生和功能恢复作用优于剂量为2 mg、12 mg组。提示GsRg1促神经再生作用的最佳用药剂量为4~8 mg·kg。②GsRg1的促进受损神经功能恢复的作用与NGF因子密切相关。

The Experimental Study on Mechanism underlying the Promoted Regenera-tion of Injured Sciatic Nerve by Ginsenoside Rg1
Abstract:Objective To observe whether the Rg1 can promote nerve function recovery after sciatic nerve injury and to in-vestigate whether the partial release nerve growth factor participate in its mechanism, on the basis of which we can make a further study of its mechanism. Methods We use male Wistar rats for making a sciatic nerve injury model. february age, a total of 270, were randomly divided into 6 group, each group of 45. Under anesthesia, we amputate the right sciatic nerve of the rats and then stitch it up immediately under the surgical microscope, taking the left sided sciatic nerve as the blank control. After the surgery, They are saline group, GsRg1 2 mg group, Gs 4 mg group, Gs 8 mg group, Gs 12 mg group and Mecobalamin group. After 2 weeks, 4 weeks and 8 weeks of surgery, we can weigh the Rat two-side triceps surae muscle measuring its wet weight ratio, we can evaluate the regeneration and functional recovery of the sciatic nerve; and detect the changes of NGF expression of sciatic nerve in rats with Immunohistochemistry and Western blot technology. Results①Tri-ceps surae muscle wet weight ratio, each dose group of GsRg1 and Mecobalamin group is also significantly higher than Saline group, in GsRg1 group, 4 mg group and 8 mg group are all higher than the other two groups of GsRg1, the recovery of GsRg1 2 mg group and 12 mg group are similar to nerve conduction velocity of Mecobalamin group and Triceps surae muscle wet weight ratio.② NGF Immunohistochemistry and Western blot results indicate that the NGF expression of GsRg1 groups and Mecobalamin group are higher than Saline group. The NGF expression of GsRg1 4 mg and GsRg1 8mg were more than Mecobalamin group, GsRg1 2 mg group and GsRg1 12 mg group. The NGF expression of Mecobalamin group is similar to GsRg1 2 mg group and GsRg1 12 mg group. Conclusion①GsRg1 could promote rat sciatic nerve transaction in-jury and enhance the nerve regeneration and functional recovery after surgical sutures;It suggests that the best dosage of GsRg1 for promoting nerve functional regeneration is 4~8 mg·kg. ②The function of GsRg1 for promoting the recovery of injured nerve is related to NGF factor.
