摘    要:为了解Bcl-2及Bax基因在输精管复通后大鼠生精细胞中的表达情况,作者建立了大鼠输精管结扎和复通模型,应用免疫组化法观察Bcl-2及Bax基因在大鼠输精管复通后生精细胞中的表达变化。结果显示:输精管复通组Bcl-2基因表达逐渐增强,复通后8周显著地强于输精管结扎组,到12周同假手术组;而输精管复通组的Bax基因表达无改变,与输精管结扎组无差异,都显著地强于假手术组。Bcl-2基因蛋白表达的变化说明在输精管结扎后,以及输精管复通后均发挥了抗凋亡的作用,而Bax基因蛋白表达在输精管复通前后的一致性,说明其对生精细胞凋亡作用有待进一步研究。

Abstract:The objective of this study was to explore the change of Bcl-2 and Bax gene expression in rat spermatogneic cells after vasostomy. A rat model with vasoligation and vasostomy was set up and then used for detecting the protein expre ssion of Bcl-2 and Bax gene at 4,8,12,16 weeks after vasostomy by means of immu nohistochemistry.Sixty rats were randomly distributed into three groups named vas oligation group(VG),vasostomy group(VOG) and shamoperation group(SOG) respecti vely. The rusults showed that the Bcl2 protein expression in VOG group increas ed gradually and came to be significantly higher than that in VG group 8 weeks af ter vasostomy, and it's level was the same as that in SOG group at the 12th week .The bax protein expressions in VG and VOG groups were significantly higher than that in SOG group, but there was no difference between VOG and VG groups. In co nclusion, the change of Bcl-2 protein expression observed in this study is ind cative of the anti-apoptosis function of Bcl-2 gene in both the status of bein g vasoligated and vasostomized, whereas the consistent Bax protein expression befo re and aften the repatency of the vas suggests further researches into its effec t on apoptosis of spermatogenic cells.
