醋酸棉酚对大鼠成熟卵泡的颗粒细胞产生雌二醇及孕酮功能影响的离体观察

摘    要:给30天幼龄大鼠皮下注射12IU 孕马血清,48小时后采集发育成熟的卵泡颗粒细胞进行离体培养,并在培养基中加入不同浓度的醋酸棉酚,以观察醋酸棉酚对卵巢甾体激素合成功能的影响。结果发现,棉酚对颗粒细胞分泌孕酮的能力以及对hCG 促进颗粒细胞生成孕酮的能力均有抑制作用。这种抑制作用随棉酚剂量的增加及作用时间的延长而增强。但同样剂量的棉酚对颗粒细胞利用睾酮转化为雌二醇的能力无明显抑制作用。这提示棉酚对颗粒细胞内孕酮生物合成系统有直接抑制作用,而对芳香化酶的活性似无明显干扰作用,表明棉酚对卵巢内甾体激素合成功能的抑制具有选择性。此外,本实验还发现,hCG 的作用在未被完全阻断时,能不同程度地削弱棉酚对颗粒细胞分泌孕酮功能的抑制作用。

Abstract:The effect of various concentrations of gossypol on ovarian steroidogenesis was studiedin granulosal cell cultured isolated from follicles of 30-day-rat after treatment with pregnantmare's serum gonadotropins.The results show that gossypol inhibited both basal and HCG-stimulated progesteronesynthesis by the granulosal cells in a dose(10~50 ug/ml)and time(4~24h)dependentmanner;but had no significant effect on estradiol production when testosterone was addedto the culture medium.These data presented an evidence for a direct interference withthe progesterone biosynthesis system in granulosal cells by gossypol.But data did not showgossypol had a significant effect on aromatase function indicating a selective inhibitory effectof it on ovarian steroidogenesis.In addition,it was found that when the effect of HCG on progesterone synthesis wasnot completely blocked by gossypol,addition of HCG to the medium,could reduce theinhibitory effect of it on progesterone secretion.
