小鼠肝癌(HCa)两株克隆细胞系转移表型的比较

摘    要:来自同一祖先的两株不同亚克隆细胞系Hca-16A3-F(F)和HCa-A2-P(P),在两种不同小鼠体内经不同途径接种所得的转移率和转移部位显示有差别:C3/he小鼠皮下组,F呈现高淋巴道转移(80.0%),而P未出现转移,C3H/he小鼠尾静脉组,F在卵巢(28.6%)和淋巴结(28.6%)有较低的转移率,P权出现一例(14.3%)卵巢转移,然而,在裸鼠体内却出现了异常的变化,F呈现高血道(肺脏71.4%)转移而不出现淋巴道和其他部位转移,但P却出现高淋巴道转移(85.7%)兼有低血道(肺脏28.6%,卵巢14.3%,肾脏28.6%)转移。体内实验结果提示:HCa细胞中含有转移、浸润能力不同的亚群;F细胞在体内具有较强的浸润力与转移力,且有淋巴道转移的倾向;P细胞的浸润力和转移力则低于F细胞,但若给其提供适当的到达器官的通路,完全能形成实验性转移灶。C3H/he小鼠也是HCa细胞生长和转移的理想宿主。

Abstract:HCa-F and HCa-P cell lines are two clones isolated from HCa cell line.After intravenous and subcutaneous tumor-cell inoculation,the different sites and rate of metastasis by HCa-F and HCa-P cell were seen in two recipient mice.HCa-F resulted in highly lymphatic metastasis(80%) in C3H/hej mice with subcutaneous injection in contrast, HCa-P was free from metastasis;in C3H/hej mice with intravenous inoculation,HCa-F cell produced 28.6% colonies in ovary and 28.6% colonies in LN, HCa-P cell cnly showed 14.3% metastasis in ovary.In athymic mice inoculated from tail vein,unusual experimental metastsis appeared.HCa-F cell formed high lung metastasis(71.4%) not accompanied by other organs involved,recipient suffered extensive metastasis by HCa-P(LN 85.7%;lung 28.6%;ovary 14.3%; hidney 28.6%).The results from in vivo experiments suggest that thare are different with difffering the capacity of invasion and metastasis.in HCa cells;HCa-F cell line show higher potential of invasion and metastasis in vivo and affinity to lymphatic system;HCa-P cell line is a cell line with much lower invasive and metastatic ability than HCa-F cell line,but they can form experimental metastasis if provided with vascular access to an organ;C3H/hej mice can be host for cells to be transplanted and metastasiszed.
