大黄对慢性便秘模型大鼠结肠血管活性肠肽水平及肠动力影响研究

摘    要:目的:采用洛哌丁胺灌胃建立便秘大鼠模型,观察大黄对便秘模型大鼠血管活性肠肽(vasoactive intestinal peptide,VIP)水平的影响。方法:健康Wistar大鼠18只,随机分为3组,空白组、模型组、大黄组。模型组及大黄组采用配制好的含洛哌丁胺(1.5 mg·kg~(-1)·d~(-1))混悬液2 m L灌胃7 d。模型建立后,大黄组以大黄(5 mg/kg)混悬液2 m L灌胃。检测结肠传输,ELISA检测AQP3含量。结果:(1)造模后大鼠的体重增加、结肠存留粪便增多、粪便含水量降低、肠道传输时间延长(P<0.05);(2)大黄干预组大鼠大便量、粪便含水量增加(P<0.05);(3)大黄干预便秘大鼠,其结肠传输时间缩短,VIP水平明显低于模型组(P<0.05)。结论:大黄可加快肠道传输,可能与降低大鼠结肠中VIP水平,促进肠蠕动有关。

Abstract:Objective:To observe the effect of Rhubarb on the vasoactive intestinal peptide (VIP)in colon tissues in the Lop-ersmide-induced constipated rats.Methods:18 healthy Wistar rats were randomly divided into 3 groups,the control group,model group and rhubarb group.Model group and rhubarb group were prepared with the fittest dosage of Lopersmide(1 .5 mg·kg -1 ·d -1 )mixed suspension 2 mL lavage 7 d.After model establishment,rhubarb group with rhubarb (5 mg /kg)mixed suspension 2 mL lavage.Detec-tion of colonic transmission,ELISA detection of AQP3 content.Results:(1 )After building the model,the rats’weight gain,the colon feces increase,waste water content reduce and the transmission time of the intestinal tract prolonged(P <0.05 );2)The feces and the waste water content of rats in rhubarb group were increased obviously(P <0.05 ).(3)Rhubarb intervention in rats with constipation, the colonic transit time was shortened,VIP level was significantly lower than the model group (P <0.05 ).Conclusion:Rhubarb can accelerate intestinal transmission,may be related to reducing the level of VIP in the colon of rats,and promoting the intestinal peristal-sis.
