流式细胞仪多参数分析白血病免疫表型

摘    要:用流式细胞仪5种参数和单克隆抗体(McAh)12项指标,对105例急性白血病(AL)思考骨髓进行免疫学分型。先用3种McAb为一组染色,区分AL的主要系列:CD_(13)/CD_(33)急性髓系白血病(AML),CD_(19)-B淋巴细胞白血病,CD_7-T淋巴细胞白血病。然后进一步用各种AL亚型相关的McAb分析抗原表达,AML表现为低分化抗原表达型,分化抗原表达丰富型及异常分化抗原表达型。AL各亚型在流式细胞仪上的表现特征有助于了解白血病细胞恶性增殖和分化的程度,对白血病预后的判断有意义。

Abstract:one marrows of 105 cases of acute leukerma (AL)were immunophenotyped with flow cytometry with 5parameters and 12 monoclonal antibodies. The first 3McAbs were used to discriminate main AL lineages:CD_(13)/CD_(33)-myeloid lineage (AML) , CD_(19)-B lymphoidlineage (B-ALL) , CD_7-T lymphoid lineage (T-ALL).Then, the cells were labeled by lineage related McAbsfor further subtyping leukemias. Samples were analysedon FACScan with a Paint-a-Gate soft ware. AML cellsshowed three patterns: differentiation antigen lowexpression, differentiation antigen high expression, andaberrant antigen expression. All showed differentiationantigens coexpression. The results suggested that theAL immunologic characters on flow cytometry could beused in studying the malignant charactersitics of AL andprognosing AL.
