健脾、补肾方药对老龄大鼠骨质疏松防治作用的实验研究

目的　评价与比较健脾、补肾及健脾补肾方药对老龄大鼠骨质疏松的防治效果。方法　以补中益气汤和右归饮分别为健脾法和补肾法的代表方药 ,选用 1 2月龄大鼠 ,随机分为 7组 :模型组 ,健脾大、小剂量组 ,补肾大、小剂量组和健脾补肾大、小剂量组。各组大鼠在同等喂养条件下分别灌胃给药 ,用药 1 0 0d后 ,测定大鼠血清Ca、P、ALP、ACP和尿HOP ,并观察骨密度及生物力学指标。结果　健脾、补肾方药及两者合用均能有效防治老龄大鼠骨量的丢失 ,用药组全身骨密度及腰椎骨密度与模型组比较差异有显著性 (P <0 0 5 ,P <0 0 1 ) ,同时能增加骨灰重及股骨最大扭曲弯度 ,增强股骨承受最大力度 ,其作用以健脾与补肾方药合用最为显著 ,用药后各组血清ALP、ACP和尿HOP水平均明显下降 ,血清Ca、P水平均明显升高 ,提示骨吸收减少及机体对Ca、P的摄入增加 ,其作用以补肾方药相对优于健脾方药 ,并以两者合用效果显著。结论　健脾、补肾方药在防治老龄大鼠骨质疏松中均有确切疗效 ,防治效果以健脾、补肾方药合用最优

Effects of spleen-strengthening and kidney-tonifying traditional Chinese herbal medicine on osteoporosis in aged rats

Objective To evaluate the effects of spleen-s tren gthening and kidney-tonifying traditional Chinese medinal herbs on the osteop orosis in aged rats Methods Buzhong Yiqi decoction was used a s a spleen-strengthening medicine, and Yougui decoction as a kidney-tonifying me dicine A total of 80 rats, 12 months old rats were randomly divided into 7 gro ups: model group, high dose and low dose of Buzhong Yiqi groups, high dose and low d ose of Yougui groups, and high dose and low dose of Buzhong Yiqi plus Yongui gro ups The medicine was administered by gavage One hundred days later,the level s of calcium, phosphorus, alkaline phosphatase (ALP) and acid phosphatase (ACP) in serum and hydroxyproline (HOP) in urine were measured The bone mineral densit y (BMD) and biomechanical parameters were also determined simultaneously Results Both Buzhong Yiqi and Yougui decoctions were effective in pr e venting the loss of bone mineral in aged rats The BMD of body and the maximal w eight bearing of femur were significantly increased in the treatment groups (P <0 05, P< 0 01) The levels of serum ALP and ACP and urine HOP wer e remar kably decreased in the treatment groups, while the levels of serum calcium and p hosphorus were evidently increased, indicating that there were more intakes of c alcium and phosphorus by gastrointestinal tract in the treatment groups than tha t in the control groups Youqui decoction was more effective than Buzhong Yiqi decoction and the greatest efficacy was evidenced groups of combined of both Conclusion Spleen-strengthening and kidney-tonifying traditio nal Chinese medicines are efficacious in prevention and treatment of osteoporos is in aged rats, and most effective by combined use of both
