反相高效液相色谱法测定治糜康栓中苦参碱和氧化苦参碱的含量

摘    要:目的建立治糜康栓中苦参碱和氧化苦参碱的含量测定方法,以期更全面地控制治糜康栓的质量。方法采用(RP-HPLC法)以十八烷基硅烷键合硅胶(C18)色谱柱(4.6 mm×250 mm,5μm),以乙腈-磷酸盐缓冲液(7:93,磷酸二氢钾1.7 g加500 mL纯水溶解,用磷酸调节pH=3)为流动相,检测波长220 nm,流速1 mL/min,分别对市售治糜康栓与实验室自制治糜康栓中苦参碱及氧化苦参碱的总含量进行测定。结果市售治糜康栓中苦参碱及氧化苦参碱的总含量约为1.52 mg/粒,实验室自制治糜康栓中苦参碱及氧化苦参碱的总含量约为2.53 mg/粒。结论建立的反相高效液相色谱方法适用于治糜康栓中苦参碱及氧化苦参碱的含量测定。

Abstract:Objective To establish a method for content determination of matrine and oxymatrine in Zhimi Kangshuan, in order to more comprehensive quality control of the Zhimi Kangshuan. Methods Octadecylsilane chemically bonded silica (C18) column (4.6×250 mm, 5 μm) was added to the acetonitrile-phosphate buffer as mobile phase (7:93, 1.7 g and 500 mL potassium dihydrogen phosphate water solution, adjusted with phosphoric acid pH=3), the detection wavelength was 220 nm, the flow rate was 1 mL/min, and the total content of matrine and oxymatrine was prepared by the preparation of the Chinese Pharmacopoeia and the self-made. Results The total content of matrine and oxymatrine in the market drugs is about 1.52 mg/li. The total content of matrine and oxymatrine in the lab made products is about 2.53 mg/li. Conclusion This paper establishes to be applicable to the determination of matrine and oxymatrine content in Zhimi Kangshuan by reversed phase high performance liquid chromatography.
