黄精提取液不同作用时间对离体蛙心收缩活动的影响

摘    要:目的 观察黄精提取液对离体蛙心不同作用时间收缩力和心率的效应.方法 制备30只离体灌流蛙心,随机分为对照组、高浓度组及低浓度组,每组10只.在灌流液中,正常对照组加入0.85%NaCl注射液,低浓度和高浓度组分别加入不同浓度黄精提取液(0.25 g·ml-1、0.5 g·ml-1)50山,通过BL-410生物机能实验系统记录加药前、加药后30 s和2min、4min、6min的心收缩力和心率,并进行比较分析.结果 黄精低浓度和高浓度组加药后2min、4min、6min心收缩力均明显低于正常对照组(P<0.01),2min心收缩力明显低于30 s心收缩力(P<0.01),而各时间段的心率与正常对照组比较差异无统计学意义(P>0.05).结论 黄精具有降低离体蛙心收缩力作用,但对心率无明显影响.

Abstract:Objective To observe the effect of polygonatum extraction on contractility and heart rate of isolated frog heart at different function time.Methods 30 isolated frog hearts in vitro were prepared,and they were randomly divided into control group, high concentration group and low concentration group with 10 in each group.In the normal control group ,the perfusate was added with 0.85 % NaCl injection,and different concentration of polygonatum extraction (0.25 g. ml-1 ,0.5 g · ml-1 ) 50 μl were respectively added to the perfusate of the high concentration group and low concentration group.The cardiac contractility and heart rate at 30 s,2 min,4 min and 6 rain before and after adding medicine,which were recorded by BL-410 biological function experiment system, were compared and analyzed.Results The cardiac contractility at 2 min, 4 min, and 6 min after adding medicine in the high and the low concentration group were significantly lower than those of the normal control group ( P 〈0. 01 ), and the cardiac contractility of 2 rain after adding medicine were significantly lower than that of 30s in the high concentration and low group( P 〈0. 01 ).There was no statistically significant difference between the heart rate of the high and low concentration group and that of normal control group in different periods( P 〉0. 05) .Conclusion Polygonatum can reduce the contractility of isolated frog heart, but there is no significant influence on heart rate.
