PPARα激动剂对高糖刺激的肾系膜细胞PEDF和VEGF mRNA表达的影响

摘    要:目的:研究色素上皮细胞衍生因子(PEDF)和血管内皮生长因子(VEGF)在糖尿病肾病中的作用以及PPARα激动剂与糖尿病肾病的关系。方法:将培养的大鼠HBZY-1肾小球系膜细胞株分为6组,高糖作为刺激因子,PPARα激动剂WY14643作为干预因素。分别设正常组,高糖组,高糖加WY14643治疗组(包括不同的WY14643浓度)以及高糖加WY14643溶剂对照组。用实时定量PCR法测定各组系膜细胞PEDF以及VEGF mRNA表达。结果:(1)与正常组相比较,高糖组肾系膜细胞PEDF mRNA的表达明显降低,而WY14643可逆转高糖导致的肾系膜细胞PEDF mRNA表达的降低。(2)高糖组肾系膜细胞VEGF mRNA的表达较正常组明显增加,而WY14643可减少高糖导致的肾系膜细胞VEGF mRNA表达的增加。结论:高糖可抑制大鼠肾系膜细胞PEDF并增加VEGF的表达,而PPARα激动剂可在一定程度上逆转高糖导致的这种改变。

Abstract:Objective: In order to explore the mechanism of pigment epithelium-derived factor (PEDF)and vascular endothelial growth factor(VEGF) in diabetic nephropathy and the relation between PPAR-alpha agonist and diabetic nephropathy, the effect of high glucose and PPAR-alpha agonist WY14643 on the expression of PEDF and VEGF in cultured rat glomerular mesangial cells and discuss the relation between PPAR-alpha agonist and diabetic nephropathy were investigated. To find new evidence for prevention and cure of diabetic nephropathy. Methods: Cultured HBZY-1 rat glomerular mesangial cells were divided into 6 groups, glucose was a stimulated factor, and PPAR-alpha agonist WY14643 was an intervenor: Normal Glucose group, High Glucose-treated group, therapy groups: High Glucose and WY14643 (including different concerntrations of WY14643), impregnant control group : High Glucose and impregnant of WY14643 group. The expression of PEDF and VEGF mRNA was measured by Real time Quantitative PCR. Results: (1)Normal Glucose group glomerular mesangial cells have expression of PEDF and VEGF, the expression of PEDF mRNA decreased and the expression of VEGF mRNA was increased in high glucose group(P<0.05). (2)The expression of PEDF mRNA in therapy groups increased greater than that in high glucoses group and the expression of VEGF mRNA in therapy groups decreased greater than that in high glucose group(P<0.05). WY14643 can inhibit the decreased expression of PEDF and the increased expression of VEGF induced by high glucose. Conclusions: High glucose can decrease significantly the expression of PEDF as well as increase the expression of VEGF in cultured mesangial cells. PPAR-alpha agonist can prevent this change induced by high glucose in cultured mesangial cells.
