摘    要:【目的】探讨亚慢性甲苯染毒对雌性sD大鼠垂体促性腺激素的影响,为阐明甲苯对雌性生殖内分泌系统的不良影响提供实验依据。【方法】60d雌性sD大鼠,随机分为4组,动情周期2周后分别给予0、35、70、330mg/kg甲苯连续灌胃染毒40~45d,每天上午阴道涂片观察动情周期的变化。于动情期处死大鼠分别提取脑垂体做免疫组化,同时测定血清中FSH和LH含量,观察甲苯对垂体促性腺激素的影响。【结果】高剂量组与对照组相比,动情周期持续时间延长、动情周期异常率升高,规则动情周期数降低,差异有统计学意义(P〈0.05)。免疫组化结果:高剂量组、中剂量组与对照组相比,棕色颗粒明显增多,两组平均光密度(MOD)和面数密度(SD)存在显著性差异(P〈0.01),低剂量组可见少量棕色颗粒,表明LH和FSH有少量表达,与对照组无明显统计学差别(P〉O.05)。血清中FSH和LH浓度结果:高剂量组与对照组相比明显增高(P〈0.05);中剂量组和对照组相比明显增高(P〈0.05);低剂量组和对照组相比无统计学差异(P〉0.05)。【结论】甲苯可引起大鼠动情周期的改变:引起大鼠垂体促性腺激素细胞分泌激素增多,同时可引起血清中FSH和LH含量的增高。

Abstract:[Objective] To analyze the effect of subchronic toluene intoxication on pituitary gonadotropin hormone through animal experiment provide an experimental evidence for the adverse effect of toluene on the genital endocrine system of female SD rats. [Methods]60-day female SD rats are randomly divided into four groups. The estrons cycle of each group is observed for two weeks. Then the rats are poisoned continually for 40 to 45 days by injecting toluene at 0 mg/kg, 35 mg/kg, 70 mg/kg and 330 mg/kg respectively. The estrous cycles' changes should be observed every morning by a vagina swear. In order to observe the influence of toluene on pituitary gonadotropin hornome, the SD rats were killed during their estrus and the pituitary gland was extracted for making a pituitary immunohistochemistry comparison, meanwhile the Follicle Stimulating Hormone (FSH) and the Luteinizing Hormone (LH) content in the serum should be measured. [ Results] The estrous cycle of high-dose group lasted longer, became irregular and the abnormal rate increased as compared with the control group. The difference was significant statistically (P 〈 0.05). Immunochemieal results revealled an obvious increase compared of brownish particles in the high and middle group. The mean optical density (MOD) and surface-number density (SD) between the high and middle dose group with significant difference (P 〈 0.01). A few brownish particles could be found in the low-dose group indicated that there was some impression of LH and FSH. The results of FSH and LH concentrations in the serum of the high-dose group was significantly higher than that of the contrast group. The differences have a statistical significance (P 〈 0.05). The result of the FSH and LH concentrations in serum of the middle-dose group was significantly higher than the one of the contrast group. The differences have a statistical significance (P 〈 0.05). Comparing the low-dose group withthe contrast group, the results of the FSH and LH concentrations in the Serum have no statistical differences (P 〉 0.05). [Conclusion] Toluene may cause the change of estrous cycle, increase the secretion of pituitary gonadotropic hormone and increase the serumcontent of FSH and LH.
