百合鳞叶总DNA提取方法的改进

摘    要:目的建立一种适合RAPD分析的百合鳞叶总DNA提取方法。方法在传统CTAB提取方法上,对其进行改良,进一步优化提取方法。结果用改良CTAB法提取的总DNA的OD260/OD280在1.6-2.0之间,电泳条带清晰,其含量和纯度完全满足RAPD分析的要求。结论该方法提取百合鳞叶总DNA简便实用。

Abstract:Objective To excogitate a method for extracting DNA from Bulbus Lilii to be appropriate for RAPD analysis.Methods Based the traditional CTAB,the method was improved to obtain an extrcting method.Results The OD260/OD280 of total DNA was between 1.6 to 2.0 with the improved method of CTAB,the banding patterns was clear,and the content and pureness of the total DNA can meet the reaction for RAPD.Conclusions A simple and useful method for DNA extraction from lily'squama leaves are created.
