摘    要:目的:建立痛风颗粒中绿原酸和阿魏酸同时测定的方法。方法:采用高效液相色谱法同时测定绿原酸和阿魏酸的含量。色谱柱:YMCJ’sphere ODS-H80 C18柱(4.6mm×150mm,5μm)。流动相:乙腈-1%冰醋酸梯度。流速:1mL/min。测定波长为324nm。结果:绿原酸与阿魏酸分别在12.5~200μg/mL和5~80μg/mL内呈良好线性关系,r均为0.9999。绿原酸平均回收率为102.48%,阿魏酸平均回收率为96.92%,RSD分别为1.25%和1.74%(n=6)。结论:本方法简便、准确,可用于痛风颗粒的质量控制。

Abstract:Objective: To establish a method for simultaneous determination of chlorogenic acid and ferlic acid in Gout Granule. Methods: High performance liquid chromatorgraphy (HPLC) was used to determine the content of chlorogenic acid and ferlic acid simultaneously. The two acids were separated on YMC J' sphere ODS-H80 C18 column (4.6 mm × 150 mm, 5 μm) and were eluted gradiently in mobile phase (acetonitrile-1% glacial acetic acid). Flow rate was set at 1 mL/min and wavelength was 324 nm.Results: The linearities were in the range of 12.5 - 200 μg for chlorogenic acid and 5 ~ 80 μg/mL for ferlic acid (r=0.999 9). The average recovery rate was 102.48% (RSD=1.25%) for chlorogenic acid and 96.92% (RSD=1.74%) for ferlic acid.Conclusion: The method is simple and accurate. It can he used for quality control of Gout Granule.
