子癎前期母血中胎儿游离DNA实时荧光定量PCR研究

摘    要:目的对子痫前期产妇母血中胎儿游离DNA进行定量测定,探讨转运的胎儿游离DNA与子痫前期的关系及其在子痫前期疾病的发展和预测中的价值。方法通过实时荧光定量PCR技术利用特异性Y性别决定区(SRY)基因对妊娠分娩男婴的52例母血浆中胎儿游离DNA进行定量测定。子痫前期产妇27例(实验组),其中轻度者12例,重度者15例;正常妊娠产妇(对照组)25例。结果52例母血浆中均检测出胎儿游离DNA,实验组浓度均值较对照组增高(P〈0.001),子癍前期重度者较轻度者增高(P〈0.001)。结论妊娠高血压子痫前期转运的母血中胎儿游离DNA浓度高于正常妊娠者,可能与胎盘滋养层和合体滋养层细胞凋亡增加有关。妊娠母血中胎儿游离DNA浓度与子痫前期具有相关性,可为作为预测和监测子痫前期的临床指标。

Abstract:Objective The free fetal DNA in preeeclampsia maternal plasm has been quantitatively assayed by real-time fluorescence quantitative PCR (FQ-PCR) to explore the correlation between the fetal DNA and preeclampsia as well as the assay's value in assessment of development and prognosis of preeeclampsia. Methods The fatal plasma DNA was assayed with real-time FQ-PCR by detecting sex-determining region Y(SRY) in the genes. A total of 52 pregnant women were selected in the study,including 12 patients of the mild degree of preeclampsia and 15 patients of the severe degree of preeclampsia. The control group included 25 normal pregnancies. Results The fetal DNA was detected in 100% of maternal plasma (52/52). The mean DNA level in the severe preeclampsia group and were significantly higher than that in the mild group(P〈0. 001). The fetal DNA level in preeclampsia was markedly higher than that in the normal pregnancy group (P〈0. 001). Conclusions T.he higher level of fetal DNA in preeclampsia maternal plasm might be correlated with the increasing apoptosis of trophoblastic cells in placenta. The free fetal DNA level in preeclampsia maternal plasm is correlated with preeclampsia which can serve as a clinical indication for diagnosing and monitoring preeclampsia.
