贯叶金丝桃总黄酮纯化工艺研究

摘    要:研究碱化法结合大孔树脂法纯化贯叶连翘总黄酮的工艺条件。方法:以静态饱和吸附量、静态解析率为指标对6种大孔树脂进行了筛选;又以总黄酮含量为指标对提取液上柱前的预处理方式、树脂吸附工艺参数进行了优选。结果:D101大孔树脂吸附量、解析率最好;上样液浓度为0.2 g生药/mL;上样速度为2BV/h;洗脱剂为60%乙醇;洗脱速度为3BV/h;洗脱剂用量为5BV/h。结论:优选的贯叶金丝桃总黄酮纯化工艺稳定、简便可行、适合大生产。

Abstract:Objective:To study the purification process conditions of total flavonids from Hypericum Perforatum with alkalization method combined with macroporous resin method. Methods : In static saturated adsorption capacity,the static analytic rate was as an index of 6 kinds of macroporous resin for selection. Total flavonoids content was as index to extract on column pretreatment method, and resin adsorption process parameters were optimized. Results :The results showed that D101 macroporous resin adsorption was the best. The sample liquid concentration was 0. 2 g/mL and the washing was 60% ethanol. The elution rate was 3 BV/ h. The dosage of eluent was 5 BV/h. Conclusion: All above showed that the optimization of total flavonids from Hypericum Perforaturn purification process is stable, simple and feasible, and suitable for mass production.
