熊果酸对人肝癌SMMC-7721细胞增殖作用的实验研究

摘    要:目的:评定熊果酸对肝癌细胞SMMC-7721的增殖作用,流式细胞术研究对肝癌细胞凋亡的影响。方法:将不同浓度的熊果酸分别与人肝癌细胞SMMC-7721共同培养24,72 h后,采用中性红染色法测定吸光度(A),评定熊果酸对肝癌细胞的增殖作用,流式细胞术研究其对肝癌细胞的凋亡。结果:熊果酸对SMMC-7721细胞增殖有明显的抑制作用(P<0.05或P<0.01),呈时间-剂量依赖关系,24 h和72 h半数抑制浓度(IC50)分别为12.59,8.32μg·mL-1;对肝癌细胞凋亡率为14.07%,药物浓度与凋亡率呈正相关。结论:熊果酸对肝癌细胞SMMC-7721增殖有明显抑制作用,该抑制作用可能与诱导肝癌细胞凋亡有关。

Abstract:Objective :To evaluate the function of ursolic acid on the proliferation of human Hepatoma carcinoma cells SMMC -7721. And to study the effect of ursolic acid on the apoptosis of SMMC -7721 by flow cytometry. Methods:Human hepatoma SMMC - 7721 cells were co - cultured with different doses of ursolic acid for 24 h,72 h, respectively. The absorbance value was e- valuated by neutral red staining method. The influence of ursolic acid on the proliferation of hepatoma cells was evaluated ; and the apoptosis was detected by using flow cytometry. Results. Ursolic acid on the proliferation of SMMC - 7721 cells had obvious inhibi- tion effect( P 〈0. 05 or P 〈0. 01 )and presented time-dose related manner. The half inhibitory concentration(IC50) of ursolic acid on SMMC -7721 cells strain was 12.59 μg · mL-l and 8.32 μg · mL-t, respectively. The result indicated that the apoptosis rate was up to 14.07% and there was a positive correlation between the drug concentration and the apoptosis rate. Conclusion:Ursolic acid on the proliferation of hepatoma SMMC -7721 cell line has the obvious inhibitory action. The inhibition may be related to the induction of ar~ootosis of heDatoma ceils.
