大鼠脊髓急性损伤后内皮素-1含量的变化

摘    要:目的:通过检测大鼠脊髓急性损伤后内皮素-1(ET-1)的含量变化,探讨ET-1在脊髓继发性损伤中可能的作用机制。方法:用35g负荷压迫大鼠胸段脊髓(T8-9)5min,造成大鼠胸段脊髓急性损伤,用放射免疫法测定大鼠脊髓急性损伤后早期血浆和伤段脊髓组织中ET-1的含量。结果:脊髓损伤后30min,血浆,伤段脊髓组织中ET-1水平显升高,4h达最高峰,并分别持续至48h和72h。结论:ET-31参与脊髓继发性损伤,可能是伤后4-24h重要的损害因子之一。

Abstract:Objective:To explore the possible role of endothelin 1(ET 1) in secondary spinal cord injury (SCI) by measuring the changes of ET 1 levels after SCI in rats. Methods:The thoracic spinal cord of rats was compressed by a load of 35 g for 5 min, causing a moderate injury. The changes of ET 1 levels in plasma and injured spinal cord tissues were examined by means of radioimmunoassay. Results:ET 1 concentrations in plasma and injured tissues were significantly increased 30 min after spinal cord compression and reached its peak at 4 h, then it restored slowly to normal level in 72 h. Conclusion:ET 1 contributes to the pathophysiological actions of secondary SCI, it may play a key role in secondary injury at 4 24 h after acute SCI.
