乙型肝炎组织中Fas及FasL表达的研究

为研究乙型肝炎肝组织中Fas及Fas配体（FasL）的表达与肝细胞损伤的关系，我们以兔抗-Fas及兔抗-FasL多克隆抗体，采用免疫组织化学分别标记和双标记技术，检测60例急性轻型、慢性活动型、活动性肝硬化患者肝组织中Fas及FasL。结果显示：Fas及FasL的检出率分别为76．7％及70．0％，Fas在肝细胞浆内表达，FasL多见于肝组织中浸润的淋巴细胞内，也见于肝细胞胞浆表达，其阳性淋巴细胞主要分布于汇管区。FasL阳性肝细胞分布与Fas肝细胞分布相同。免疫组化双标记染色示Fas及FasL可在同一肝细胞胞浆内表达。提示Fas／FasL系统在乙型病毒性肝炎肝细胞损伤中起重要作用。

Study on the Expression of Fas and Fas Ligand in Liver Tissue of Patients with Hepatitis B

To explore the expression of liver cell injury and their relationship of Fas and Fas ligand in livertissue of patients with hepatitis B infection, 60 liver biopsy or autopsy specimens including 20 cases of acute mildhepatitis, chronic active hepatitis, and active cirrhosis were studied by immunohistochemical double labeling tech-nique by using polyclonal anti-Fas and anti-Fas ligand. The detection rate for Fas and Fas ligand were 76. 7% (46/60) and 70. 0% (42/60) respectively. Fas antigen was located in cytoplasm of hepatocytes. Fas ligand expressedmainly in infiltrating lymphocytes in portal or periportal areas, and also in some cytoplasm of hepatocytes. Thedistribution of Fas ligand-positive hepatocytse was similar to those of Fas-positive hepatocytes in liver tissue. Dou-ble labeling studies showed that both Fas and Fas ligand expressed in the same hepatocyte. These findings suggestFas/Fas ligand system may play an important role in liver cell injury by hepatitis B virus infection.
