摘    要:应用高效液相色谱法测定人体血浆、尿液中莫雷西嗪及其亚砜、砜代谢产物的生物浓度,本法选用国产硅胶YWG-C18为固定要,甲醇、乙腈、水、三乙胺为流动相,检测波长254nm.采用离子对一步提取法提取样品,莫雷西嗪,莫雷西嗪亚砜和莫雷西嗪亚砜和莫西嗪的最低血浆检测浓度为30、8、10nm/gml,绝对回收率大于85%。

Abstract:A rapid, specific and sensitive method using HPLC for the simultaneous determination of moricizine and its two metabolites in plasma and urine was developed. The compounds were extracted from biologic samples using a single ion-pair extraction.The residue was redissolved in 0.1 of 0. 01mol/L glacial acetic acid and separated on a C18 reversed-phase column. The mobile phase consisted of methanol,acetonitrile,water and triethylamine (pH 5.4)with 254nm UV detector. The lowest detection limits of moricizine, moricizine sulphoxide and moricizine sulphone plasma concentration were 30,8 and 10ng/ml,respectively. The absolute recoveries of moricizine and its two metabolites were all over 85%,The utility of the method for human pharmacokinetic studies was demonstrated.
