紧密连接蛋白claudin 1、3、4与肺爆震伤致大鼠肺水肿的关系研究

摘    要:目的 探讨大鼠肺泡上皮细胞紧密连接蛋白claudin 1、3、4与肺爆震伤致肺水肿的关系。方法 应用爆炸装置制备大鼠肺爆震伤模型,伤后4h留取标本,通过测定支气管肺泡灌洗液中蛋白含量及肺组织湿/干重比值评价肺水肿,电镜下观察肺泡上皮对镧的扩散,肺泡上皮基膜染色,通过免疫组织化学技术及计算机图像分析评价肺组织中claudin 1、3、4蛋白的表达。结果 肺爆震伤发生4h后肺水肿形成,肺泡上皮间基膜断裂,镧颗粒大量进入肺泡上皮细胞间,肺组织中claudin 1、3、4蛋白表达相较于对照组显著降低(P<0.05)。结论 肺爆震伤发生后,紧密连接蛋白claudin 1、3、4表达下降,紧密连接受损,肺泡上皮细胞屏障被破坏,从而导致肺水肿的形成。

Abstract:Objective To discuss the relationship between rat alveolar epithelial cell tight junction proteins claudin 1,3,4 and pulmonary edema after blast lung injury. Methods Blast lung injury model was made with explosive device, and specimens were taken after four hours. By measuring the protein in bronchial alveolar lavage fluid and wet/dry ratio,we evaluated pulmonary edema. We used TEM to observe the diffusing of lanthanum between alveolar epithelial cell. Claudin 1,3,4 expression in lung tissue by was evaluated.Alveolar epithelial basement membrane staining and through immunohistochemistry and computer image analysis. Result Pulmonary edema occurred after blast lung injury, and alveolar epithelial basement membrane ruptured and a large number of lanthanum ran into alveolar epithelial cells interval. There was significant decrease in the expression of claudin 1,3,4 on alveolar epithelial cells, as compared with control group(P<0.05). Conclusion After blast lung injury,the expression of claudin 1,3,4 were decreased, tight junction was impaired, alveolar epithelial cells barrier was destroyed and induced pulmonary edema.
