姜黄素对溃疡性结肠炎大鼠模型的疗效评价及其作用机制

摘    要:目的观察姜黄素作用于大鼠溃疡性结肠炎模型的疗效及其机制。方法选用雄性健康SD大鼠60只,随机分成4组,每组15只。A组:正常对照组;B组:模型组;C组:姜黄素治疗组;D组:地塞米松治疗组。TNBS灌肠后,按DAI的标准评分,进行大体损伤评分,HE染色后组织损伤评分,记录每日的进食、进水量及死亡数。测定结肠组织中丙二醛(MDA)及谷胱甘肽过氧化物酶(GsH-Px)、超氧化物歧化酶(SOD)和髓过氧化物酶(MPO)的活性。结果C组大鼠的病情改善、进食量和进水量均优于B、D组,死亡率低于B组与D组。DAI评分、大体损伤形态、HE染色评分均显示C、D组分别均与B组比较差异有极显著性(P<0.01),C、D组比较差异有显著性(P<0.05)。MDA和MPO活性在B组明显升高,并高于D组和C组(P<0.05),而D组低于C组(P<0.05)。SOD和GsH-Px在B组明显降低,C、D组明显高于B组(P<0.05),C组高于D组(P<0.05)。结论姜黄素是治疗溃疡性结肠炎大鼠模型的有效药物。姜黄素的疗效比地塞米松为优。姜黄素可能是通过降低肠组织中的MDA、MPO的活性和升高SOD、GsH-Px的活性达到抗炎目的。

Abstract:Objective Ulcerative colitis rat model was induced by 2,4,6-trinitrobenzene sulfonic acid(TNBs) and treated by curcumin,comparing to the curative effect of Dexamethasone,the mechanisms of curcumin treating colitis rat model were studied on the base of the upper.Methods 60 male Sprague-Dawley rats were ran-domly divided into 4 groups,Group A:the normal group,Group B: the model group,Group C: curcumin group,Group D: Dexamethasone group.We observed and noted the symptoms of all the rats in every group,including avoirdupois,properties of stool and blood in stool,grading on the standard of DAI,at the same time,noting the quantity of taking food and water,and mortality every day.We graded histopathologic and macroscopical changes.The activity of colon MDA,GSH-Px,SOD and MPO in all rats was also detected,respectively.Results Curcu-min could significantly improve symptom,reduced colon histopathologic and macroscopical injury.Its curative ef-fect was superior to Dexamethasone.Curcumin decreased the content of colon MDA and MPO,but the effect was feeble compaed to Dexamethasone.Curcumin could also enhance the activity of GSH-Px and SOD,and was powerful than Dexamethasone.Curcumin increased the quantity of taking food and water,decreased mortality.Conclusion Curcumin has positive effect in treating TNBs-induced colitis in rats.The curative effect of curcumin is superior to Dexamethasone.Curcumin reduces colonic inflammation through decreasing the content of colon MDA,MPO and enhancing the activity of GSH-Px,SOD.
