西洋参茎叶总皂苷对急性心肌梗死大鼠心肌能量代谢的影响

摘    要:目的探讨西洋参茎叶总皂苷是否能改善缺血心肌细胞的能量代谢。方法选用急性心肌梗死模型大鼠,随机分为心悦胶囊大、中、小剂量组,倍他乐克组,模型组,同时设假手术及正常组(每组各8只)。药物治疗组于术后第2天开始灌胃给药,每日1次,术后第15天取心肌组织制成匀桨,采用高效液相色谱(HPLC)法测定心肌组织中ATP、ADP、AMP的含量,并计算总腺苷酸(TAN)含量及能荷(EC)值。结果心悦胶囊治疗组ATP的含量及EC水平均高于模型组,其中心悦胶囊大剂量组ATP的含量及心悦胶囊大、中、小剂量组的EC水平升高显著(P<0.05)。结论西洋参茎叶总皂苷能提高缺血心肌组织中ATP含量及EC的贮备水平。

Abstract:Objective To investigate whether Panax quinquefolium saponin(PQS) could improve ischemic myocardial energy metabolism in rats with acute myocardial infarction(AMI).Methods Wistar rats with AMI were randomly divided into 5 groups(n=8): large dose of PQS group,middle dose of PQS group,low dose of PQS group,metoprolol group and model group.At the same time,sham operation and normal groups were established.Rats of treatment groups received PQS or metoprolol respectively by intragastric administration once per day,and other groups received same volume of saline.The contents of adenosine triphosphate(ATP),adenosine diphosphate(ADP) and adenosine monophosphate(AMP) in the myocardial homogenate were determined by HPLC on 15th day,and the values of total adenine nucleotides(TAN) and energy charge(EC) were calculated. Results The contents of ATP in large dose of PQS group and the levels of EC in PQS groups were greater than those in model group(P<0.05).Conclusion PQS can elevate the content of ATP and the level of EC in the ischemic myocardium of rats with AMI.
