营养干预对大鼠视网膜Fos蛋白和羧基肽酶E表达的影响

摘    要:目的 : 探讨牛磺酸和多种微量营养素能否通过调节光照和暗适应条件下 Fos和羧基肽酶 E( CPE)蛋白表达 ,进而影响视功能尤其是暗适应功能。方法 :  Wistar大鼠随机分为三组 ,即对照组 (正常饲料组 ) ,实验 1组 ( 5倍需要量组 )和实验 2组 ( 1 0倍需要量组 ) ,喂养 4w后 ,每组动物再随机分为光照组和暗适应组 (平均照度为 3.0 3lx) ,以正常饲料喂养 72 h,大鼠活杀取样做免疫组织化学染色。结果 : 光照和暗适应条件下 CPE和 Fos蛋白在视网膜各层次细胞中均有广泛分布 ,且光照和暗适应条件下两者表达无明显差异 ,其中 Fos蛋白主要集中分布于内核层、内网状层和节细胞层 ,大鼠补充一定剂量牛磺酸和微量营养素后 ,可使光照时外网状层、内核层和节细胞层 c- fos基因表达增强 ,而对光照时 c- fos基因表达无明显影响。结论 : 牛磺酸和多种微量营养素干预可使光照或暗适应条件下 CPE和 Fos蛋白表达和分布发生明显变化 ,尤其是暗适应条件下 c- fos基因表达的变化可能对视杆通路视觉信号传递产生重要的调节作用。

Abstract:Objective: To investigate the molecular mechanism of the influence of taurine and micronutrients on visual signal transmission.Methods: Wistar rats were divided into three groups: control group, experiment group 1 and experiment group 2, and fed for 4 weeks on normal diet, diets containing 5 times and 10 times requirements of taurine, vitamin A,B 1,B 2,PP, zinc and selenium of rat respectively. Then each treatment group was divided into light group and dark adaptation subgroups. After feeding normal diet for another 3 days in different environments, all animals were killed and the expression of CPE and Fos protein were analysed in retina.Results: The CPE and Fos protein were widely distributed in different retinal cell types in dark and light environments, and Fos protein was expressed predominantly in outer plexiform layer, inner plexiform layer and ganglion cell layer. The CPE expression in outer plexiform layer, inner nuclear layer in light and outer plexiform layer in dark, the Fos expression in outer plexiform layer, inner plexiform layer and ganglion cell layer in dark adaptation were increased after treatment of taurine and micronutrients. Conclusion: The expression and distribution of CPE and Fos protein in retina were quite different in dark and light environments after intervention of taurine and other micronutrients, which may be one of molecular mechanisms of vision improvement.
