不同IL-15基因转染对 NCI-H446肿瘤细胞增殖的影响

摘    要:目的探讨不同形式IL-15基因转染对人小细胞肺癌细胞株NCI—H446细胞增殖的影响。方法用本室已构建的3种转染不同IL-15基因(原型、成熟肽及以IL-2信号肽替代IL-15信号肽的改型IL-15基因)的NCI.H446细胞模型:CIL-15、CIL-15mp和CIL-2sp-IL-15mp,以野生株NCI-H446细胞(CW)为对照,台盼蓝拒染计数法测定细胞生长曲线、MTT法检测细胞增殖、流式细胞术分析细胞周期的变化、RT-PCR法检测cyclin D1和cyclin E的表达。结果与CW相比,CIL-15、CIL-15mp和CIL-2sp-IL-15mp增殖速率依次减慢、G0/G1期细胞比例依次增加、S期细胞和细胞周期蛋白cyclin E表达依次减少、cyclin D1表达无变化。结论3种IL-15基因转染均有使NCI—H446细胞增殖明显减慢的作用,强度依次为改型〉成熟肽〉原型;这种作用与降低NCI-H446细胞cyclinE的表达有关(r=0.953,P〈0.05),与cyclin D1的表达无关(r=0.155,P〉0.05)。

Abstract:Objective To investigate the effects of the transfections of different IL-15 genes on NCI-H446 cell proliferation. Me- thods By using three kinds of NCI-H446 cell models transfected respectively by the IL-15 prototype gene, the IL-15 mature peptide gene, and the IL-15 modified gene that the signal peptide of IL-15 was replaced with that of IL-2 (their names were C_ IL-15 , C_ IL-15mp , and C_ IL-2sp-IL-15mp , respectively) as experimental groups, and the wide NCI-H446 cells (C_W) as control group, the cell proliferations were measured by trypan blue exclusion and MTT, the distributions of cell cycle was analyzed by flow cytometry, and the mRNA expressions of cyclin D1 and cyclin E were detecteded by semi-quantitive RT-PCR. Results Compared with C_W, in the three kinds of cell models, the cell proliferations were decreased, the cell percentages were increased at G0/G1 phase and decreased at S phase, and the mRNA expression was decreased in cyclin E but not changed in cyclin D1. Conclusion The three kinds of IL-15 genes, especially the IL-15 modified gene, can decrease the proliferation of NCI-H446 cells, which may related to decreasing the expression of cyclin E.
