升气壮阳汤超声雾化法对变应性鼻炎小鼠的作用研究

摘    要:目的探讨升气壮阳汤超声雾化法对变应性鼻炎小鼠的干预作用及可能作用机制。方法选取SPF级BALB/c小鼠40只,随机取10只作为空白组,其余小鼠采用卵清蛋白+氢氧化铝免疫辅助剂构建变应性鼻炎小鼠模型,将成模小鼠随机分为模型组、氯雷他定组、升气壮阳汤组,每组10只。氯雷他定组给予氯雷他定药液灌胃,升气壮阳汤组给予升气壮阳汤超声雾化,空白组和模型组给予生理盐水灌胃,均连续干预28 d。记录各组小鼠造模前及干预第7,14,28天鼻部症状评分,干预结束后检测各组小鼠血清免疫球蛋白E(IgE)、组胺、白细胞介素-4(IL-4)、白细胞介素-2(IL-2)、白细胞介素-5(IL-5)、白细胞介素-10(IL-10)、γ-干扰素(IFN-γ)水平,实时荧光定量PCR法检测各组小鼠鼻黏膜组织中GATA3、T-bet、Tim1、Tim3 mRNA表达量。结果干预第7,14,28天,模型组鼻部症状评分均明显高于空白组(P均<0.05),氯雷他定组和升气壮阳汤组均明显低于模型组(P均<0.05)。模型组血清IgE、组胺、IL-4、IL-5、IL-10水平及鼻黏膜组织中GAT3、Tim1、Tim3 mRNA表达量均明显高于空白组(P均<0.05),血清IL-2、IFN-γ水平及鼻黏膜组织中T-bet mRNA表达量均明显低于空白组(P均<0.05);氯雷他定组和升气壮阳汤组血清IgE、组胺、IL-4、IL-5、IL-10水平及鼻黏膜组织中GAT3、Tim1、Tim3 mRNA表达量均明显低于模型组(P均<0.05),血清IL-2、IFN-γ水平及鼻黏膜组织中T-bet mRNA表达量均明显高于模型组(P均<0.05)。结论升气壮阳汤超声雾化可减轻变应性鼻炎小鼠的症状,其作用机制可能与降低炎症因子表达,调控Th1/Th2动态平衡有关。

Abstract:Objective It is to explore the effect of ultrasonic nebulization of Shengqi Zhuangyang Decoction on allergic rhinitis mice and its possible mechanism.Methods Forty SPF BALB/c mice were randomly selected,in which 10 ones as the blank group,and the rest mice were used to establish allergic rhinitis mouse models by ovalbumin+aluminum hydroxide immunization adjuvant,and the successful models were randomly divided into model group,loratadine group,Shengqi Zhuangyang Decoction group,10 rats in each group.The loratadine group was given intragastric administration of loratadine,the Shengqi Zhuangyang Decoction group was given ultrasound atomization of Shengqi Zhuangyang Decoction,the blank group and the model group were given normal saline by intragastric administration,and the intervention was continuous for 28 days.The nasal symptom scores of the mice in each group were recorded before the modeling and on the 7 th,14 th,and 28th day of the intervention.After the intervention,the levels of serum immunoglobulin E(IgE),histamine,and interleukin-4(IL-4),interleukin-2(IL-2),interleukin-5(IL-5),interleukin-10(IL-10),interferon-γ(IFN-γ),real-time fluorescence quantitative PCR method was used to detect the expression of GATA3,T-bet,Tim1,and Tim3 mRNA in the nasal mucosal tissues of mice in each group.Results On the 7,14,and 28 day of intervention,the nasal symptom scores of the model group were significantly higher than those of the blank group(all P<0.05),and the scores in the loratadine group and Shengqi Zhuangyang Decoction group were significantly lower than those of the model group(all P<0.05);the levels of serum IgE,histamine,IL-4,IL-5,and IL-10 and the expression of GAT3,Tim1 and Tim3 mRNA in the nasal mucosa in the model group were significantly higher than those in the blank group(all P<0.05),and the levels of serum IL-2,IFN-γand T-bet mRNA expression in nasal mucosa tissues were significantly lower than those in the blank group(all P<0.05);the levels of serum IgE,histamine,IL-4,IL-5,IL-10 and the expression of GAT3,Tim1,and Tim3 mRNA in the nasal mucosa in the loratadine group and Shengqi Zhuangyang Decoction group were significantly lower than those in the model group(all P<0.05),the levels of serum IL-2 and IFN-γand the expression of T-bet mRNA in the nasal mucosa were significantly higher than those of the model group(all P<0.05).Conclusion Ultrasonic atomization of Shengqi Zhuangyang Decoction can alleviate the symptoms of allergic rhinitis mice,and its mechanism may be related to reducing the expression of inflammatory factors and regulating the dynamic balance of Th1/Th2.
