施今墨对药配方对2型糖尿病大鼠氧化应激的影响

摘    要:目的观察施今墨对药配方对2型糖尿病大鼠血糖、游离脂肪酸(FFA)和氧化应激的影响,初步探讨其作用机制。方法随机选取6只Wistar大鼠作为正常组,余44只采用高脂高糖饲养+腹腔注射链脲佐菌素(35 mg/kg)方法制备2型糖尿病大鼠模型,将造模成功大鼠随机分为模型组、二甲双胍组及施今墨对药配方高、中、低剂量组,二甲双胍组给予1 g/kg二甲双胍灌胃,施今墨对药配方高、中、低剂量组给予23.2 g/kg、11.6 g/kg、5.8 g/kg施今墨对药配方灌胃,正常组和模型组灌胃等量生理盐水,均连用8周。干预8周后检测大鼠体重、空腹血糖(FPG)、餐后2 h血糖(2hPG)及血清C肽、超氧化物歧化酶(SOD)、FFA、丙二醛(MDA)水平,计算胰岛素抵抗指数(HOMA-IR)。结果与正常组比较,模型组大鼠FPG、2hPG、FFA、MDA水平和HOMA-IR均明显升高(P均<0.05),大鼠体重和血清SOD、C肽水平均明显降低(P均<0.05)。与模型组比较,二甲双胍组、施今墨对药配方各组大鼠血清SOD、C肽水平和施今墨对药配方低剂量组大鼠体重均明显升高(P均<0.05),二甲双胍组和施今墨对药配方中、低剂量组FPG、2hPG、FFA、MDA水平及施今墨对药配方低剂量组HOMA-IR均明显降低(P均<0.05)。结论施今墨对药配方可以减缓2型糖尿病大鼠体重下降情况,降低血糖水平,其机制可能与抑制氧化应激、改善胰岛素抵抗有关。

Effect of Shijinmo herbal-pair formula on oxidative stress in type 2 diabetic rats

Abstract:Objective It is to observe the effect of Shijinmo herbal-pair formula on blood glucose,free fatty acid(FFA)and oxidative stress,and explore its mechanism.Methods Six Wistar rats were randomly selected as the normal group,and the remaining 44 rats were fed with high fat and high sugar diet+intraperitoneal injection of streptozotocin(35 mg/kg)to prepare type 2 diabetic rat models.The successful rat models were randomly divided model group,metformin group and the high,medium and low dose groups of Shijinmo herbal-pair decoction,the metformin group was given 1 g/kg metformin by gavage,and the high,medium and low dose groups of Shijinmo herbal-pair decoction were given 23.2 g/kg,11.6 g/kg,5.8 g/kg Shijinmo herbal-pair decoction by gavage,the normal group and the model group were given the same amount of normal saline,all the groups were treated for 8 weeks.After 8 weeks of intervention,the rats’body mass,the levels of fasting fasting plasma glucose(FPG),2 h postprandial blood glucose(2hPG)and serum C peptide,superoxide dismutase(SOD),FFA,and malondialdehyde(MDA)were measured,and insulin resistance index(HOMA-IR)was calculated.Results Compared with the normal group,the levels of FPG,2hPG,FFA,MDA and HOMA-IR in the model group were significantly increased(P<0.05),while the body weight and the levels of serum SOD and C peptide were significantly decreased(P<0.05).Compared with the model group,the levels of serum SOD and C peptide in metformin group and the high,medium and low dose groups of Shijinmo herbal-pair decoction and the body weight in the low dose group were significantly increased(P<0.05),the levels of FPG,2hPG,FFA and MDA in metformin group and the medium and low dose groups of Shijinmo herbal-pair decoction and HOMA-IR in the low dose group were significantly decreased(P<0.05).Conclusion Shijinmo herbal-pair formula could alleviate the weight loss and reduce the blood glucose level of type 2 diabetic rats.The mechanism may be related to the inhibition of oxidative stress and the improvement of insulin resistance.
