扁桃体和外周血CD8+T细胞表达CXCR5和IL-21水平对比分析

目的 对比分析扁桃体和外周血单个核细胞(PBMC8)中CD8+T细胞表达CXCR5和IL-21的水平是否存在差异.方法 分离扁桃体和正常人PBMCs,四色流式细胞术(FCM)检测扁桃体单个核细胞和PBMCs中CD8+T细胞表达CD69和CXCR5 的水平.采用佛波酯(PMA)和离子霉素(Ionomycin)刺激扁桃体单个核细胞和PBMCs后,流式细胞术检测CD8+T细胞表达IL-21的水平.结果 与PBMCs中CD8+T细胞相比,扁桃体组织中CD8+T细胞高表达CD69和CXCR5(P＜0.05),经PMA+Ionomycin刺激后,扁桃体CD8+T细胞可检测到IL-21的表达,而PBMCs中CD8+T细胞基本不产生IL-21.结论 CD8+T细胞的表型和功能在扁桃体组织和PBMCs中存在差异,扁桃体中CD8+CXCR5+和CD8+IL-21+T细胞的免疫功能有待进一步研究.

Comparative analysis of the expression of CXCR5 and IL-21 on CD8+ T cells from tonsil and peripheral blood

Objective To evaluate the differences of CXCR5 and IL-21 expression on CD8+ T cells from tonsil and peripheral blood.Methods The tonsillar mononuclear cells and peripheral blood mononuclear cells(PBMCs) were isolated,then four color flow cytometry(FCM) analysis was used to compare the expression of CD69 and CXCR5 on CD8+ T cells from tonsillar mononuclear cells and PBMCs.The expression of IL-21 on CD8+ T cells was also tested after stimulated by PMA and Ionomycin.Results The expression of CD69 and CXCR5 was significantly higher on CD8+ T cells from tonsil compared with that on CD8+ T cells from PBMCs(P<0.05).The expression of IL-21 was also detectable on CD8+ T cells from tonsil after stimulated by PMA and Ionomycin.In contrast,CD8+ T cells from PBMCs did not produce IL-21.Conclusion The phenotype and function of CD8+ T cells from tonsil and PBMCs are different,the immunologic function of CD8+CXCR5+ and CD8+IL-21+ T cells from tonsil need to be studied in depth.
