督脉配穴对染铅幼年大鼠海马一氧化氮合酶阳性细胞的保护作用

摘    要:目的探讨督脉配穴对染铅幼年大鼠海马不同亚区一氧化氮合酶(NOS)变化的拮抗作用。方法将40只SD大鼠随机分为3组,即对照组、染铅组、督脉配穴组。采用反映学习记忆功能的Y迷宫法测试大鼠神经行为的改变,用NADPH-黄递酶(NADPH.d)组织化学法研究大鼠海马不同亚区NOS的活性神经元变化。结果染铅组大鼠的学习记忆能力比督脉配穴组和对照组明显下降(P〈0.05),但督脉配穴组与对照组之间差异无统计意义(P〉0.05);组织化学实验结果显示,染铅组大鼠海马CA1区和齿状回的NOS阳性神经元明显少于督脉配穴组和对照组(P〈0.05),在CA3区无差别,督脉配穴组与对照组各亚区之间差异无统计意义(P〉0.05)。结论铅可损伤幼年大鼠学习记忆能力,其机制可能与染铅后海马各区NOS活性的不同变化有关,督脉配穴针灸治疗对铅引起的学习记忆损伤和NOS活性的变化有拮抗作用。

Abstract:Objective To investigate the antagonistic effect of governor meridian acupuncture on the nitric ox- ide synthase level in young lead-exposed rat hippocampus subarea. Methods 40 SD rats were randomly divided into three groups, normal group, lead-exposed model group and governor meridian acupuncture group. Y-maze test was applied to study the effects of lead on learning and memory ability in rats; NADPH-d histochemistry was used to investigate the changes of nitric oxide synthase (NOS) in different hippocampal subareas in lead-exposed young rat. Results Compared with the control and governor meridian acupuncture group, the learning and memory ability of lead-exposed rats was significantly decreased (P〈0.05), but there was no statistic significance between governor meridian acupuncture group and control group (P〉0.05) ; The number of NOS positive neurons on CA1 and dentate gyrus of lead-exposed young rats were significantly decreased ( P〈0.05 ), but no significant changes in CA3 ; No difference between the control and governor meridian acupuncture groups in CA1, CA3 and dentate gyrus ( P〉0.05 ). Conclusion Lead may damage the learning-memory ability of the young rats, which might be related to the changes of NOS in different subfields of hippocampus in lead-exposed rats, governor meridian acupuncture might play an important role in preventing lead-inducted damages.
