摘    要:目的:普伐他汀是一种HMGCoA还原酶的竞争性抑制剂,通过抑制胆固醇的合成降低血中胆固醇的含量。建立一种灵敏可靠的测定大鼠肌肉中普伐他汀含量的方法。方法:以固相提取技术从肌肉匀浆中提取富集普伐他汀,采用高效液相色谱法紫外检测测定。结果:最低检测限0.4ng,定量限5ng/g肌肉,方法线性范围5~100ng/g肌肉,固相提取回收率52%~66%,方法精密度为6%~18%。结论:建立了灵敏、准确测定大鼠肌肉中普伐他汀的方法,并测定了该药经灌胃后在肌肉组织中的含量。

Abstract:Objective: To develop a sensitive and reliable method to determine pravastatin in rat muscle. Methods: Pravastatin was extracted from muscle homogenate with the technique of solid phase extraction and then was analyzed by RPHPLC with UVdetector. Results: The lowest detection limit of pravastatin was 0.4 ng and the quantitation limit was 5 ng/g of muscles(RSD 20%).The linearity range was 5100 ng/g of muscle and the solid phase extraction recovery was 5266%.The precision(RSD) of the method was 618%. Conclusion: The HPLC procedure is sensitive with good precision to assay pravastatin in rat muscle.A pharmacokinetic application to determine pravastatin in muscle had been practiced.
