高效液相色谱法测定大鼠肌肉中普伐他汀的含量

目的：普伐他汀是一种ＨＭＧＣｏＡ还原酶的竞争性抑制剂，通过抑制胆固醇的合成降低血中胆固醇的含量。建立一种灵敏可靠的测定大鼠肌肉中普伐他汀含量的方法。方法：以固相提取技术从肌肉匀浆中提取富集普伐他汀，采用高效液相色谱法紫外检测测定。结果：最低检测限０．４ｎｇ，定量限５ｎｇ／ｇ肌肉，方法线性范围５～１００ｎｇ／ｇ肌肉，固相提取回收率５２％～６６％，方法精密度为６％～１８％。结论：建立了灵敏、准确测定大鼠肌肉中普伐他汀的方法，并测定了该药经灌胃后在肌肉组织中的含量。

Determination of Pravastatin in Rat Muscle by RPHPLC

Objective: To develop a sensitive and reliable method to determine pravastatin in rat muscle. Methods: Pravastatin was extracted from muscle homogenate with the technique of solid phase extraction and then was analyzed by RPHPLC with UVdetector. Results: The lowest detection limit of pravastatin was 0.4 ng and the quantitation limit was 5 ng/g of muscles(RSD 20%).The linearity range was 5100 ng/g of muscle and the solid phase extraction recovery was 5266%.The precision(RSD) of the method was 618%. Conclusion: The HPLC procedure is sensitive with good precision to assay pravastatin in rat muscle.A pharmacokinetic application to determine pravastatin in muscle had been practiced.
