苯妥英钠对创伤性脑组织单胺类神经递质变化的影响

摘    要:目的探讨苯妥英钠(DPH)对大鼠颅脑创伤后脑组织单胺递质变化的影响。方法将60只大鼠随机分为对照组、脑创伤组和DPH干预组。用自制撞击器撞击大鼠颅顶,复制出闭合性颅脑创伤。分别于损伤1h和6h后应用荧光分光法测定脑组织匀浆单胺神经递质含量。结果与对照组比较,创伤组在脑损伤1h后,多巴胺(DA)、5-羟色胺(5-HT)、去甲肾上腺素(NE)无明显变化;在脑损伤6h后,脑单胺神经递质的含量显著升高(P<0.05);DPH组在脑损伤1h、6h后DA、5-HT显著升高(P<0.05),NE无明显变化。与创伤组比较,DPH组在脑损伤1h、6h后5-HT、DA、NE均低,但无明显差别。结论DPH有助于提高5-HT等单胺类神经递质含量,通过改善通气状况对缺血、缺氧性脑创伤具有保护作用。

Abstract:Objective To probe the effect of diphenylhyduntoin(DPH) on monoamine neurotransmitters induced by traumatic brain injury (TBI). Methods The sixty Wistar rats were divided into control group ,TBI group, DPH group randomly. The top of skull of the rats was attacked by self-made bio-impact machine and a model of closed craniacerebral injury was established. To the experimental DPH(40 mg/kg) was given before TBI. The contents of dopamine(DA), norepinephrine(NE) and 5-hydroxytryamine(5-HT) in the brain tissues were measured by fluorescent methods. Results Compared with control group, TBI group of DA, 5-HT, the NE has no significant differenceat 1 hour; the monoamine neurotransmitters of brain were significant increased (P<0.05) at 6 hours. DPH group of DA,5-HT were significant increase (P<0.05), NE has no significant difference at 1 and 6 hours. The TBI group and DPH group comparison, the 5-HT, DA and NE of DPH group at the 1 and 6 hours is all low in the TBI group, but has no significant difference.Conclusion The DPH can raise the content of monoamine neurotransmitters. The blood lacking and anoxia brain wound can be protected by improving ventilate condition.
