川芎嗪对心肌细胞缺氧/复氧损伤的保护作用

摘    要:目的 研究川芎嗪对培养的SD乳鼠心肌细胞缺氧/复氧(H/R)损伤的保护作用及其机制.方法 将原代培养的SD乳鼠心肌细胞按随机数字表法分为5组,分别为正常组、H/R损伤模型组和H/R+川芎嗪给药组(川芎嗪浓度分别为50、100、150 μg·mL-1),每组10孔.测定各组心肌细胞超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量及乳酸脱氢酶(LDH)活性.结果 与H/R损伤模型组相比,川芎嗪给药组可明显提高SOD活性,降低MDA含量、LDH活性,差异均有统计学意义(P<0.05).结论 川芎嗪对体外培养心肌细胞模拟H/R损伤模型有明显的保护作用,其保护机制可能与川芎嗪抑制自由基生成或清除自由基有关.

Abstract:Objective To investigate the protective effects of ligustrazine on hypoxia/reoxygenation injury and its mechanisms of action in cultured SD neonatal rat cardiomyocytes.Methods The primary cultured cardiomyocytes were randomly divided into five groups:normal group,hypoxia/reoxygenation model group,50 μg·mL-1ligustrazine group,100 μg·mL-1ligustrazine group and 150 μg·mL-1ligustrazine group,with 10 wells in each group.Superoxide dismutase(SOD) activity,malondialdehyde(MDA) concentrations and lactate dehydrogenase(LDH) content were measured.Results Ligustrazine significantly induced a increase in SOD activity and a decrease in MDA concentrations and LDH content,as compared to hypoxia/reoxygenation model group(P0.05).Conclusion Ligustrazine may protect cultured neonatal rat cardiomyocytes from hypoxia/reoxygenation injury through a mechanism involving inhibiting and scavenging free radicals.
