人胰岛素样生长因子-I 对高血压病人肠系膜动脉平滑肌细胞增殖周期的影响

摘    要:目的在细胞水平研究高血压病人动脉血管平滑肌细胞(ASMC)的生长特性和体外血管活性物质对ASMC增殖作用的影响。方法应用复合酶法进行高血压病人ASMC分离,并用人胰岛素样生长因子I(HILGF-I)干预以观察人ASMC的增殖情况。结果经低浓度的HILGF-I作用,高血压病人ASMC呈典型的“谷”与“峰”样生长,经历了10多个月传代培养至35代。发现该细胞生长分裂活性活跃,与人胎儿脐动脉增殖活性相似。结论复合酶法能使高血压病人的ASMC的静止期缩短,传代后生长特征稳定性强,高血压病人体外培养ASMC的增殖有赖于HILGF-1的存在。

Abstract:Aim In order to study the proliferation characters of human mesentric artery smooth muscle cells(ASMC) and the effects of activating substances in blood in vitro,the authers investigated human ASMC with the interference of human insulin like growth factor 1(HILGF 1). Methods With the compound enzymetic method the authers have isolated the human hypertensive mesentric arterial smooth muscle cells successfully. They studied the effects of HILGF 1 on ASMC proliferation characters in vitro. Results With a low concentration of HILGF 1 theyfound human ASMC showing a typical peak and valley growth figure after 10 months culture under the interfering of PMA. The growing character of 35th passage DMC kept a stable living character under the interference of the HILGF 1. The proliferation of infantal ASMC depended on the HILGF 1 concentration, but the hypertensive ASMC not. HILGF 1 can increase the DNA synthesis obviously. Also HILGF 1 can promote the hypertensive ASMC get into the S and G 2 M periods. Conclusion Also treated with HILGF 1 on human ASMC,this method can shorten the ASMC isolating shock period, a stable living figure and purified at the same time as ASMCs isolated from host's vessels. In addition, the growth of human hypertensive ASMC were deprived on the existing of HILGF 1 in culture.
