猫局灶脑缺血模型的改进并建立功能检查模型

摘    要:目的 对猫持续性局灶性脑缺血模型加以改进 ,使其制作简便 ,模型标准、稳定 ,并建立功能检查模型。方法 猫麻醉后 ,经眶后暴露大脑中动脉 ,在其发出外侧纹状体动脉的外侧部电凝闭塞 ,以医用耳脑胶及明胶海绵粘合硬膜和骨窗。在术前及术后以1 8FDG(1 8F fluorodeoxyglucose)作示踪剂 ,进行PET(positronemissiontomography)检查 ,经计算机处理 ,得出各脑区的标准吸收值 (thestandardizeduptakevalue ,SUV)及脑结构图像。术后第 7天处死动物 ,行TTC染色 ,确定梗死部位及计算体积。结果 手术后在猫大脑中动脉 (MCA)皮层分布区均产生梗死灶 ,7d后的梗死体积为 (19 3± 0 70 ) % ,动物无死亡及并发症的发生 ;猫脑PET图像大体结构清晰 ,MCA闭塞后 1h即可见该侧MCA皮层区放射性浓聚影明显减低 ,且与最后梗死区相一致。结论 该模型制作方便 ,重复性及稳定性好 ,适合于脑血管疾病的功能性研究

Abstract:Objective To improve the cat model of permanent focal cerebral ischemia and to detect functional change by 18 FDG-PET in cat.Method Focal cerebral ischemia was produced in 5 cats by permanent occlusion of the middle cerebral artery(MCA) via the transorbital approach.The occlusion site was located lateral to the origin of the lateral striate artery.The dural and orbital bone defects were covered with pieces of gel foam and ear-brain glue.Before and after MCAO,the 18 FDG-PET method was used to detect the cerebral ischemia change.Seven days after MCAO,the cats were sacrificed for HE stain,and the volume.Manfarction was calculated.Results The model was evaluated by neurological scoring and TTC staining.Manifestations of cerebral infarction were found in all cats,and the volume of infarction was about (19.3±0.70)%.There was no death and complication in all cats.With 18 FDG-PET examination,the picture of cerebral structure was clearly demonstrated and the standardized uptake value(SUV) of left Sylvian's gyrus was lower than that of the right.Furthermore,the area of decreased SUV was in well accordance with the infarction area.Conclusion The model has excellent stability and reproducibility and is suitable for experimental studies of cerebral ischemia,especially for cerebral functional study.
