脾虚大鼠模型脑内精氨酸加压素水平和基因表达变化及归脾汤的影响

摘    要:目的观察脾虚模型脑内对学习记忆有促进作用的精氨酸加压素(AVP)水平和基因表达变化及归脾汤的影响作用。方法采用苦降泻下、饮食失节和劳倦过度法建立脾虚大鼠模型,以免疫组化和原位杂交法检测下丘脑腹侧核、海马CA1区、前额叶皮层AVP水平和基因表达变化。结果模型组上述脑区AVP免疫阳性反应物水平明显降低,治疗组明显升高;模型组海马CA1区、前额叶皮层AVPmRNA表达显著下降;治疗组表达明显升高。结论脾虚模型脑内对学习记忆有促进作用的AVP水平和基因表达有变化,归脾汤可能通过调节AVP水平和基因表达而影响其学习记忆。

Abstract:Objective To observe the level changes of arginine vasopressin (AVP)(with the effect of(improving) learning and memory abilities) and gene expression in the brain of spleen-deficiency rat model and influence of Guipi Decoction.Methods To establish the rat model of spleen-deficiency by the(methods) of purgation with bittle and cold nature drugs,improper diet and overstrain.(Immunohistochemistry) and in situ hybridization were used to detect AVP level and gene expression in nucleus hypothalamicus(ventromedialis),CA1 zone of hippocampus and prefrontal cortex.Result The immunity positive reactant of AVP in above brain zones decreased obviously in the model group,while increased obviously in the treatment group.AVP mRNA expressions in CA1 zone of hippocampus and prefrontal cortex decreased obviously in the model group,while increased obviously in the treatment group.Conclusion AVP(with the effect of improving learning and memory abilities) level and gene expression in the brain of spleen-(deficiency) rat model has changed.Guipi Decoction can influence the learning and memory abilities by regulating AVP level and gene expression.
