地黄寡糖对SAMP8小鼠造血祖细胞增殖的作用

摘    要:采用造血祖细胞体外克隆培养等实验血液学技术, 研究了地黄寡糖对快速老化模型P系小鼠(SAMP8)骨髓造血祖细胞增殖作用的影响. 结果表明地黄寡糖可促进SAMP8小鼠骨髓粒系巨噬系祖细胞, 早期和晚期红系祖细胞的增殖, 其脾细胞条件液也可使造血祖细胞克隆集落数明显增加, 地黄寡糖还可使其基质细胞层上粒系巨噬系祖细胞集落的产率明显增多. 提示地黄寡糖可能通过多种途径激活机体组织, 特别是造血微环境中的某些细胞, 促进其分泌多种造血生长因子而增强造血祖细胞的增殖.

Abstract:Effect of Rehmannia glutinosa oligosaccharide (RGOS) on the proliferation of hematopoietic progenitors was studied by using the techniques of colony culture of hematopoietic progenitors in vitro. Results indicated that RGOS 10-20 mg·kg-1·d-1×8 d, ig enhanced DNA synthesis of bone marrow cells and increased the numbers of progenitors of granulocytes/macrophages and the early and the late differentiated progenitors of erythrocytes in senescence-accelerated mouse P8 subseries (SAMP8). The growth of granulocyte/macrophage colony forming unit (CFU-GM), erythroid colony forming unit (CFU-E) and erythroid burst forming unit (BFU-E) was significantly enhanced in the presence of spleen cell conditioned media and the secretion of interleukin-6 was also increased in RGOS groups. By observing the growth of CFU-GM on stromal layer (SL), which was formed by cultivating bone marrow cells, the interaction of hematopoietic progenitors and stromal cells was studied. It showed that RGOS increased the number of CFU-GM on SL. These results suggest that RGOS can strongly enhance the proliferation of hematopoietic progenitor.
