苦参碱抗肝纤维化机理的体外研究

目的 :通过苦参碱对大鼠贮脂细胞株HSC -T6增殖及胶原、透明质酸合成的影响作用 ,研究苦参碱抗肝纤维化的作用机理。方法 :采用大鼠肝贮脂细胞株 ,以细胞增殖抑制率为指标 ,用噻唑蓝 (MTT)法 ,测定药物对大鼠贮脂细胞株HSC -T6增殖的影响 ;3H -脯氨酸掺入法 ,测定药物对HSC -T6胶原合成的影响 ;放免法测定药物对HSC -T6合成透明质酸的作用。结果 :苦参碱对大鼠贮脂细胞株HSC -T6增殖 ,胶原和透明质酸合成的抑制率随药物浓度增加而升高 ,呈药物浓度依赖关系。结论 :苦参碱通过抑制贮脂细胞增殖及细胞外基质的合成 ,发挥抗肝纤维化的作用

Study on the Mechanism of Matrine on Hepatic Fibrosis in vitro

Aim To study the mechanism of matrine on hepatic fibrosis by the inhibition of proliferation of HSC-T6, synthesis of collagen and hyaluronic acid.Methods The effects of matrine on proliferation of HSC-T6 were observed by MTT colorimetric assay,synthesis of collagen was observed by the incorporation of 3 H-proline, synthesis of hyaluronic was detected by radioimmunoassay.Results Matrine inhibited the proliferation of HSC-T6,synthesis of collagen and hyaluronic acid in dose-dependent manner.Conclusion Matrine inhibited hepatic fibrosis by inhibition of HSC-T6 cell proliferation, collagen synthesis and hyaluronic acid synthesis significantly in vitro.
