小鼠上颌突和下颌突间充质细胞成骨分化的体外研究

摘    要:目的:研究上、下颌突间充质细胞在体外的成骨能力。方法 :体外培养胚胎13.5d(E13.5)的小鼠上、下颌突间充质细胞,观察其细胞形态。取第1代细胞进行成骨诱导培养7 d,通过免疫荧光、ALP染色、茜素红染色和qPCR检测其成骨能力。应用SPSS 18.0软件包对实验结果进行独立样本t检验。结果:E13.5的小鼠上、下颌突间充质细胞能在体外成功培养和传代。成骨诱导可促进上、下颌突间充质细胞表达成骨标志物,但下颌突间充质细胞成骨标志物OCN和OPN的表达、ALP染色和茜素红染色较上颌突间充质细胞弱。结论:上、下颌突间充质细胞在体外能成骨分化,下颌突间充质细胞较上颌突间充质细胞的成骨能力稍弱。

Comparation of the osteogenic differentiation of mouse maxillary primordium and mandibular primordium mesenchymal cells

Abstract:PURPOSE: To investigate the osteogenic capacity of maxillary primordium and mandibular primordium mesenchymal cells in vitro. METHODS: Maxillary primordium and mandibular primordium mesenchymal cells were obtained from E13.5 mouse embryos and cultured in vitro. Cells from the first passage were cultured in the osteogenic medium for 7 days. Then immunofluorescence staining, ALP staining, Alizarin red staining and qPCR were applied to evaluate the osteogenic capability. Comparisons of data between two groups were assessed by an independent samples test with SPSS 18.0 software package. RESULTS: The E13.5 maxillary primordium and mandibular primordium mesenchymal cells were successfully cultured and passaged in vitro. Osteogenic induction could promote the expression of osteogenic markers both in maxillary primordium and mandibular primordium mesenchymal cells, and the expression of OCN and OPN, ALP staining and Alizarin red staining were weaker in mandibular primordium mesenchymal cells than maxillary primordium mesenchymal cells. CONCLUSIONS: Both maxillary primordium and mandibular primordium mesenchymal cells could be differentiated into osteoblast, and the mandibular primordium mesenchymal cells showed weaker osteogenic capacity than maxillary primordium mesenchymal cells.
