福建口岸输入登革I型病毒E基因序列分析

摘    要:目的研究分析福建口岸输入登革I型病毒E基因序列,为登革热诊断的分子溯源提供参考。方法收集2012—2013年福建口岸截获的7例入境Ⅰ型登革热病例资料及血清标本,用C6/36细胞培养分离病毒,提取病毒RNA,用RT-nested-PCR扩增包含E基因全长的核酸片段,扩增产物经纯化、测序,用生物学软件进行DNA序列比对分析。结果用C6/36细胞从7例登革热病例血清中成功分离出7株登革病毒株,经RT-nested-PCR证实均为DENV-Ⅰ型,构建E基因全长进化树分析发现,分离株与新加坡、越南和菲律宾等地的病毒株同源性最高,与病例入境时进行的流行病学调查资料完全吻合。结论从分子水平证明登革热病例血清中分离到的毒株为DENV-Ⅰ型病毒,结合流行病学调查资料,证实均为输入性感染病例,最有可能来源于东南亚一带。

Study on E gene sequence of dengue virus type I at Fujian port

Abstract:Objective To study E gene sequence of dengue virus type I at Fujian port, and to lay a foundation for molecular diagnosis of dengue fever. Methods Epidemiological data and serum samples were collected and viruses were isolated in C6/36 cells. The viral RNA was extracted, and amplified to DNA fragment containing the full- length E gene by RT-nested-PCR. The amplified product was purified, sequenced and analyzed with biological software. Results Seven strains of dengue virus isolates from patients at Fujian port were shown to be DENV-I. The full length E gene (1485 bp) was amplified successfully. The phylogenetic tree showed more homology with Singapore, Vietnam and Philippines virus strains, consistent with the source of entry patients. Conelusion A total of 7 cases were imported dengue fever cases. Southeast Asia was the most likely source.
