异丙基肾上腺素心力衰竭大鼠模型心肌MMP-2,9的改变

摘    要:目的建立异丙基肾上腺素(ISO)诱导SD大鼠充血性心力衰竭(CHF)模型,观察心肌MMP-2,9的变化。方法大鼠皮下注射异丙基肾上腺素2次(170mg/kg),6周以左室射血分数≤45%确定造模成功,18周使用左心导管进行血流动力学检查,取心肌采用RT—PCR,real-time PCR及明胶酶谱法检测MMP-2,9的表达,并观察病理形态学特征。结果与正常对照组相比,ISO组大鼠左心室重量指数明显增加(P〈0.01)。ISO组大鼠左室/体重比率和心肌MMP-2,9的浓度都明显升高(P〈0.01)。结论应用大削量ISO诱导大鼠充血性心力衰竭模型,该作用可能与心肌MMP-2.9激活有关。

The Changes of Cardiac MMP - 2 and MMP - 9 in Rats with lsoproterenol - induced Chronic Heart Failure

Abstract:Objective To establish the animal model of isoproterenol(ISO)-induced chronic heart failure(CHF) in rats and to study the changes of cardiac matrix metalloproteinase - 2( MMP - 2)and MMP - 9. Methods sixty male Sprague - Dawley rats were randomly allocated to two groups:The ISO- induced CHF group(n= 50) receiving two subcutaneous injections of 170 mg ISO perkilogram of body and the control group(n= 10) receiving two subcutaneous injections of 0.25 mL normal saline. At 6th week,the left ventrieular ejection fraction(LVEF) were measured by echocardiogram, the remaining rats with an LVEF≤45 %. At 18th week, the MMP- 2 and MMP - 9 in cardiac were detectedby RT- PCR, realtime PCR and gelatin zymography assay method . The mass ratio of heart ventricle and left ventricle to body were measured. Results Compared with the control group,the mass ratio of left ventricle to body and the concentration of MMP - 2 and MMP - 9 increased distinctly in ISO - induced CHF group(P〈0.01). Conclusion Activation of.cardiac MMP - 2 and MMP - 9 in rats were observed in high - dose ISO - induction rat model of CHF.
