淫羊藿苷对血管平滑肌细胞周期及周期相关蛋白表达的影响

摘    要:目的 研究淫羊藿苷(icariin.ICA)对同型半胱氨酸(homocysteinemia,HCY)诱导增殖的血管平滑肌细胞(vascular smooth musckle cell.VSMC)细胞周期的影响及其机制。方法建立HCY诱导的兔VSMC增殖模型,与不同浓度的ICA共同培养48h后,用流式细胞仪检测VSMC细胞周期变化及细胞周期调控相关蛋白Cyclin D1和CDK4的表达。结果0.2mg/L、0.4mg/L ICA组G0/G1期细胞数明显增多,s期细胞数明显减少,与HCY组相比有统计学意义(P〈0.01);CyclinDl和CDK4的表达量均随ICA剂量增加而减少。结论 ICA桔抗HCY诱导的兔VSMC增殖.其主要机制可能与Cyclin D1及CDK4的低表达所致的细胞周期抑制有关。

Effect of Icariin on Cell Cycle and Cell Cycle Related Proteins of Vascular Smooth Muscle Cell

Abstract:Objective To investigate the effect and its mechanism of icariin(ICA) on the cell cycle of homocysteinemia(HCY) induced prolif- eraled vascular smooth muscle cells(VSMC). Methods To establish model of HCY induced proliferated VSMC in rabbit and test the changes of cell cycle of VSMC and cell cycle related protein CyclinDl and CDK4 by flowmetry after cohabitued with ICA in ranked concentration for ,48 hours. Results There was significanl difference in cell cycle,CyclinD1 and CDK4 expression between 0.2 mg/L and HCY group,0.4 mg/L ICA group and HCY group(P〈0.01). The cells increase in G0/G1 stage and decrease in stage S on the experimental groups while the expression of Cyclin D1 and CDK4 decrease with the ascending of the concentration. Conclusion ICA could antagonist the proliferation of the HCY induced VSMC. Its mechanism might be highly related with the CyclinD1 and CDK4 pathways.
